摘  要:通过克隆蛹虫草(Cordyceps militaris)野生型菌株Cm01的laeA基因(CmLaeA)、合成构巢曲霉(Aspergillus nidulans)的laeA基因(AsLaeA),构建不同过表达载体,利用农杆菌(Agrobacterium tumefaciens)介导转化野生型蛹虫草菌株LN9、Cm01,获得4个过表达菌株,分别命名为CmLaeALN9(CmLaeA基因转化菌株LN9)、AsLaeALN9(AsLaeA基因转化菌株LN9)、CmLaeACm01(CmLaeA基因转化菌株Cm01)、AsLaeACm01(AsLaeA基因转化菌株Cm01),结果表明:与野生型菌株相比,过表达菌株CmLaeACm01、AsLaeALN9、AsLaeACm01的虫草素含量提高(2.5倍),CmLaeACm01的分生孢子产量提高(1.5倍)、出草时间提前5~7 d。LaeA的调控作用因不同来源的LaeA和不同的宿主菌而异。The laeA genes of the wild type Cordyceps militaris Cm01(CmLaeA)and Aspergillus nidulans(AsLaeA)were cloned and synthesized,respectively,and then used to construct corresponding overexpression vectors.Using Agrobacterium tumefaciens mediated transformation,these overexpression vectors were then transferred into wild C.militaris strains Cm01 and LN9 respectively to generate four recombinant strains as follows:CmLaeALN9(transformation of LN9 with CmLaeA),CmLaeACm01(transformation of Cm01 with CmLaeA),AsLaeALN9(transformation of LN9 with AsLaeA)and AsLaeACm01(transformation of Cm01 with AsLaeA).Compared with the wild type strains,CmLaeACm01,AsLaeALN9 and AsLaeACm01 contained 1.5 times more cordycepin.CmLaeAcm01 produced 50%more conidia and started to develop fruiting bodies 5-7 d earlier than the wild type strains.The regulatory effect of LaeA varies with source and host strain.
