摘 要:初步探究白头翁汤调控溃疡性结肠炎(UC)NKT细胞表达转录因子白细胞介素-13(IL-13)的信号通路和转录因子的作用机制。实验采用白头翁汤灌胃对C57BL/6小鼠噁唑酮结肠炎模型进行治疗。根据小鼠疾病活动指数DAI、组织病理学判断白头翁汤及其阳性对照美沙拉嗪的疗效。采用HE染色观察白头翁汤对结肠黏膜NKT细胞浸润的影响,采用实时荧光定量PCR(RT-PCR)和酶联免疫吸附实验(ELISA)方法检测组织中IL-13的mRNA表达水平和mRNA浓度,采用Western blot方法检测白头翁汤对结肠黏膜NKT细胞表达转录因子IL-13的信号通路和转录因子以及对结肠上皮屏障的影响。结果显示:小鼠噁唑酮结肠炎模型构建成功后,小鼠体重出现明显的下降,DAI评分升高,经白头翁汤治疗后,小鼠体重下降减缓,并且小鼠的DAI评分降低(P<0.05);另外,白头翁汤改善了小鼠噁唑酮结肠炎模型的病理结构变化,组织损伤程度明显降低。Western blot检测发现,白头翁汤导致IL-13信号通路相关蛋白表达呈现不同程度的升高和降低,并且也改变了结肠上皮组织细胞连接蛋白的表达。HE结果印证相关蛋白表达的变化改善了结肠组织的病理结构。研究表明白头翁汤可能是通过降低溃疡性结肠炎NKT细胞表达转录因子IL-13来提高小鼠UC治疗效果,初步机制可能是激活了IL-13相关通路蛋白和结肠上皮细胞连接蛋白的表达。The purpose of this paper is to preliminarily explore the signaling pathway and mechanism of Baitouweng decoction in regulating the expression of transcription factor IL-13 in NKT cells of ulcerative colitis.In this study,the Baitouweng decoction was used to treat C57BL/6 mice colitis model with oxazolidone.Based on the disease activity index(DAI)and histopathology of mice,the study judged the curative effect of Baitouweng decoction and its positive control mesalazine.It used HE staining to observe the effect of Baitouweng decoction on the infiltration of NKT cells in colon mucosa.RT-PCR and ELISA were used to detect the level and concentration of IL-13 mRNA in tissues.Western blot method was used to detect the effect of Baitouweng decoction on the signaling pathway and transcription factor of IL-13 expression in colon NKT cells and on the epithelial barrier of colon.The weight of mice decreased significantly,and the DAI score increased after the successful construction of oxazolidone colitis model on mice.However,after the treatment of Baitouweng decoction,the weight loss of mice slowed down,and the DAI score of mice decreased(P<0.05).In addition,Baitouweng decoction improved the pathological structure of mice with oxazolidone colitis model,and the degree of tissue damage was dramatically reduced.Western blot showed that the expression of IL-13 signaling pathway related proteins increased and decreased in different layers,and which also changed the expression of connexin in colon epithelial cells.HE results showed that the expression of related proteins improved the pathological structure of colon.Baitouweng decoction may improve the therapeutic effect of UC in mice by reducing the expression of transcription factor IL-13 in NKT cells of ulcerative colitis,and the primary mechanism may be the activation of the expression of IL-13 related pathway proteins and intraepithelial cell connexin.
