摘 要:[目的]为实现对羟基苯甲酸的生物合成。[方法]通过形态学和分子学鉴定得到一株球孢白僵菌B2660,利用B2660对苯甲酸进行全细胞催化,经高效液相色谱(HPLC)和GC-MS检测确定产物为对羟基苯甲酸,并通过正交试验对全细胞催化条件进行了优化。[结果]未优化前,球孢白僵菌B2660在48 h合成对羟基苯甲酸0.434 g/L,转化率为31.4%。经过优化后B2660在30℃、pH值7和底物浓度为2 g/L的条件下,可全细胞催化产生对羟基苯甲酸1.2 g/L,转化率达到86.9%,比优化前提高了1.68倍。[结论]球孢白僵菌B2660为首次发现具有将苯甲酸物质全细胞催化合成生成对羟基苯甲酸的能力,为生物法合成对羟基苯甲酸开辟了新途径。[Objective]To realize the biosynthesis of p-hydroxybenzoic acid.[Method]Beauveria bassiana B2660 was identified through morphological and molecular evaluation methods.The B2660 whole cell was used for sodium benzoate catalyzing reaction.The product was determined p-hydroxybenzoic acid by high performance liquid chromatography(HPLC)and LC-MS.The whole cell catalysis conditions were optimized by orthogonal experiments then.[Result]Before optimazaiotn,the production of p-hydroxybenzoic acid reached 0.434 g/L within 48 hours,with 31.4%as conversion rate.After optimization,when the whole-cell catalysis was carried at the temperature of 30℃,pH value of 7,with 2 g/L sodium benzoate,to generate 1.2 g/L of p-hydroxybenzoic acid.The final conversion rate was 86.9%,which was 1.68 times higher than that before optimization.[Conclusion]Beauveria bassiana B2660 is firstly discovered for having whole-cell catalysis ability for conversion of p-hydroxybenzoic acid from benzoic acid.It provides a new way for p-hydroxybenzoic acid biological transformation.
