摘  要:为探索空间站内航天复合应激环境下模型大鼠大脑皮层形态及蛋白表达的变化,将20只大鼠随机分为对照组与航天复合应激模型组,通过尾吊、孤养、噪声、时序变化42 d建立模拟航天复合应激模型。造模成功后,将各组大鼠大脑皮层组织进行尼氏染色与抗体芯片筛选差异蛋白。结果表明:与对照组相比,模拟长期航天应激模型会造成大鼠大脑皮层神经元损伤,神经元数量显著减少(P<0.01);Mod组抗体芯片发生显著变化的蛋白有28个,其中上调的有21个,下调的有7个;KEGG通路富集分析变化较为显著的信号通路有MAPK、p53等与神经系统相关的信号通路。航天复合应激环境会导致大鼠大脑皮层脑组织形态发生改变,也会对皮层组织中相关蛋白产生影响。To explore the changes of cerebral cortex morphology and protein expression in model rats under space composite stress environment in the space station,20 rats were randomly divided into the control group(Ctrl)and the space composite stress model group(Mod).A simulated space composite stress model was established through tail suspension,orphan rearing,noise,and timing changes for 42 days.After the model was successfully established,the rat cerebral cortex tissues of each group were Nissl stained and the antibody chips were used to screen differential proteins.Compared with the control group,the simulated space composite stress model would cause neuron damage in the cerebral cortex of rats,and the number of neurons were significantly reduced(P<0.01).Twenty-eight proteins in the Mod group changed significantly,of which 21 were up-regulated and 7 were down-regulated.The results of KEGG pathway enrichment analysis suggested that the signal pathways with significant changes were MAPK,p53 and other signal pathways related to the nervous system.Researches showed that the space composite stress environment could cause changes in the morphology of rat cerebral cortex tissue and affect the changes in related proteins in the cortex tissue.
