摘  要:目的研究积雪草挥发油的神经保护活性及其化学组成。方法采用DPPH自由基清除法和Ellman法分别评价了积雪草挥发油的抗氧化和乙酰胆碱酯酶抑制活性,采用噻唑蓝比色法测定其对β-淀粉样蛋白(Aβ)和1-甲基-4-苯基-吡啶离子(MPP^(+))诱导的SH-SY5Y神经细胞损伤的影响,采用气质联用(GC-MS)技术分析其成分。结果积雪草挥发油具有较强的抗氧化和乙酰胆碱酯酶抑制活性,对SH-SY5Y细胞有明显的促生长作用,对Aβ和MPP^(+)诱导的SH-SY5Y神经细胞损伤有显著的改善作用。积雪草挥发油中含有圆柚酮、亚油酸甲酯、角鲨烯和γ-生育酚等成分。结论积雪草挥发油在治疗神经退行性疾病方面具有研究价值。OBJECTIVE To investigate the neuroprotection related activities and the chemical constituents of Centella asiatica essential oil(CAEO).METHODS Antioxidant and acetylcholinesterase inhibitory activities of CAEO were evaluated using DPPH method and Ellman method,respectively.Effects on SH-SY5 Y cells injured by Aβand MPP;were assessed using methyl thiazolyl tetrazolium colorimetric method.Gas chromatography-mass spectroscopy(GC-MS)was used to investigate its components.RESUITS The results exhibited that CAEO showed strong antioxidant activity and acetylcholinesterase inhibitory activity.It also stimulated the growth of SH-SY5 Y cells and showed significant ameliorative effect on the injury of cultured SH-SY5 Y cells induced by Aβand MPP;.CAEO contains nootkatone,linoleic acid methyl ester,squalene andγ-tocopherol,and soon.CONCLUSION CAEO is valuable to be studied and developed on the aspect of neurodegenerative diseases’treatment.
