摘 要:目的:比较早期肥胖、早期2型糖尿病(T2DM)及早期1型糖尿病(T1DM)大鼠空肠病变的差异。方法:选取SD雄性大鼠40只,随机分为4组,分别为对照组、早期肥胖组、早期T2DM组、早期T1DM组,每组10只。通过喂养高脂饲料构建肥胖大鼠模型,高脂饲料加腹腔注射小剂量链脲佐菌素(STZ,30mg/kg)构建T2DM模型,直接腹腔注射STZ(60mg/kg)构建T1DM大鼠模型,对照组注射等量生理盐水。记录各组大鼠血糖变化,取空肠制成石蜡切片,经HE和免疫组化染色,观察分析其形态结构与紧密连接蛋白ZO-1、Occludin蛋白表达。结果:早期T2DM大鼠和早期T1DM大鼠血糖升高显著。早期T2DM和早期T1DM大鼠黏膜下层明显增厚;早期肥胖大鼠ZO-1、Occludin蛋白表达增加,而早期T1DM大鼠ZO-1、Occludin蛋白表达降低。结论:早期肥胖、早期T2DM与早期T1DM大鼠空肠组织形态均发生明显病理改变,但紧密连接蛋白表达出现差异化变化,揭示不同类型代谢性疾病早期空肠的不同变化,为早期干预提供实验依据。Objective:To compare the lesions of jejunum in early obesity,early T2DM and early T1DM rats.Methods:40 male SD rats were randomly divided into four groups:control group,early obesity group,early T2DM group and early T1DM group,with 10 rats in each group.The obese rats model was constructed by feeding high-fat diet,high-fat diet plus intraperitoneal injection of small dose of streptozotocin(STZ,30mg/kg)to construct a T2DM rats model,and intraperitoneal injection of STZ(60mg/kg)to construct a T1DM rats model.During the experiment,the blood glucose of rats were recorded,and then jejunum tissue was taken and made paraffin sections.HE staining and immunohistochemical staining were performed,and the tissue morphology and the expression of ZO-1 and Occludin proteins were observed.Results:To compared with the control group,the blood glucose of early T2DM rats and early T1DM rats increased.The submucosa of early T2DM and early T1DM rats was obviously thickened,the expression of ZO-1 and Occludin increased in early obese rats,but decreased in early T1DM rats.Conclusion:Early obesity,early T2DM and early T1DM rats had obvious pathological changes in jejunum tissue morphology,the expression of tight junction protein showed differential changes,which revealed the different changes of early jejunum in different types of metabolic diseases,and provided some experimental basis for early intervention.
