摘  要:目的:通过比较正常人和乳腺癌患者乳腺组织中miRNA表达状况,寻找可以作为生物标志物的miRNA。方法:采用GEO数据库中的GSE26659、GSE44899、GES44124数据集和TCGA数据库的乳腺癌测序数据,通过差异分析和生存分析确定筛选表达差异的miRNA,评估选出miRNA的分类性能,进行靶基因预测、富集分析并构建蛋白相互作用网络。结果:通过分析乳腺癌芯片数据,筛选出3个有作为生物标志物潜力的miRNA,经TCGA数据验证,miR-96-5p、miR-425-5p对肿瘤和正常组织具有较好的区分能力。结论:miR-96-5p、miR-425-5p有作为效应标志在乳腺癌筛查发挥作用的潜力。Objective:To find miRNAs that can be used as biomarkers by comparing the miRNA expression in breast tissues of normal people and breast cancer patients.Methods:GSE26659,GSE44899,GES44124 datasets from GEO database and breast cancer sequencing data from TCGA database were used to screen differentially expressed miRNAs by difference analysis and survival analysis.The classification performance of miRNAs was evaluated and selected.Target gene prediction,enrichment analysis and protein interaction network were constructed.Results:By analyzing analysis of breast cancer chip data,three miRNAs with potential as biomarkers were screened out.Verified by TCGA data,miR-96-5p and miR-425-5p hah good ability to distinguish tumors from normal tissues.Conclusion:miR-96-5p and miR-425-5p have the potential to act as effect markers in breast cancer screening.
