高效液相法测定蜂蜜中甘油的含量

摘　　要：建立了以混合溶剂直接提取测定蜂蜜中甘油含量的方法。采用ZORBAX Carbohydrate analysis(4.6 mm×250 mm 5-Micro)色谱柱,以乙腈/水(80∶20,v/v)为流动相,示差检测器,使用乙腈/甲醇/水混合作为溶剂快速检测甘油。结果表明,应用此法的甘油浓度在10.0 mg/L~250 mg/L范围内相关系数为0.99925,线性良好,定量限为50 mg/kg;加标回收率为94.5%~102.8%,相对偏差为1.05%~2.46%。该方法具有简单、快速、灵敏度高等特点,能有效降低检测成本,可用于批量快速测定蜂蜜中甘油的含量。A method for direct extraction and determination of glycerol content in honey by mixed solvent was established.The glycerin in honey was extracted by the mixture of acetonitrile-methanol-water,and then determined by HPLC-RIDwith ZORBAX carbohydrate analysis(4.6 mm×250 mm 5-Micro)and the mobile phase of acetonitrile/water(80∶20).The results indicated that the correlation coefficient of glycerin concentration in the range of 10.0 mg/L~250 mg/L was 0.99925,showing a good linear relationship,the quantitative limit was 50 mg/kg;the recovery rate was 94.5%~102.8%and the relative deviation was 1.05%~2.46%.This method had the characteristics of simplicity,rapidity and high sensitivity,which could effectively reduce the detection cost and could be used for batch rapid determination of glycerin content in honey.
