摘 要:目的研究白藜芦醇(resveratrol, RSV)缓解脂多糖(LPS)诱导的急性肺损伤的机制。方法随机选择40只8周龄雄性ICR小鼠,将其随机分成5组,对照组、模型组、实验组(100、300、500 mg/kg),分别用生理盐水或白藜芦醇灌胃预处理2周。用4%水合氯醛麻醉小鼠,经气道滴入2.5 mg/kg LPS复制急性肺损伤模型,对照组滴加生理盐水。构建模型后6 h,处死成功诱发急性肺损伤的小鼠,取其肺组织进行肺湿重与干重(W/D)比测定,进行组织病理学观察;利用NADP;/NADPH检测试剂盒检测烟酰胺腺嘌呤二核苷酸磷酸(NADPH)氧化酶活性,利用qRT-PCR与免疫印迹法对NADPH氧化酶亚基p47phox、p22phox表达水平进行检测。结果通过气道滴入LPS成功构建急性肺损伤小鼠模型;通过病理学观察、测定肺W/D,证明白藜芦醇以剂量依赖的方式有效缓解了LPS诱导的肺损伤;与模型组相比,实验组NADP;/NADPH比值显著降低;qRT-PCR和Western blot实验结果显示,与模型组相比,白藜芦醇显著降低了NADPH氧化酶亚基p22phox、p47phox的表达水平。结论白藜芦醇通过降低NADPH氧化酶亚基p22phox、p47phox的表达水平,降低肺组织NADP;/NADPH比值,缓解LPS诱导的急性肺损伤。Objective To investigate the effect of resveratrol(RSV) on the activity of NADPH oxidase in lipopolysaccharide(LPS)-induced acute lung injury in mice.Methods Forty male ICR mice aged 8 weeks were randomly divided into five groups: control group, model group and experimental group(100、300、500 mg/kg resveratrol),which were pretreated with normal saline or resveratrol for 2 weeks.Mice were anesthetized with 4% chloral hydrate.LPS was injected into the airway at a dose of 2.5 mg/kg.Normal saline was injected into the control group.Six hours after the model of acute lung injury was established, the mice were killed and the lung tissues were taken, and the ratios of wet weight to dry weight(W/D)were recorded.NADP;/NADPH detection kit was used to detect the activity of nicotinamide adenine dinucleotide phosphate oxidase(NADPH),and qRT-PCR and Western blot were used to detect the expression of NADPH oxidase subunits p22 phox and p47 phox.Results Acute lung injury model was successfully established in mice by instillation of LPS into the airway.Through pathological observation and determination of lung W/D,it was confirmed that resveratrol could effectively alleviated LPS-induced lung injury in a dose-dependent manner.Compared with the model group, NADP;/NADPH ratio was significantly decreased in the experimental group, and the expression of NADPH oxidase subunits p22 phox and p47 phox was significantly decreased by resveratrol in qRT-PCR and Western blot.Conclusion Resveratrol can alleviate LPS-induced acute lung damage by reducing the expression level of p22 phox and p47 phox as well as the NADP;/NADPH ratio.
