摘  要:为了评估不同浓度下甲基炔诺酮(norgestrel,NGT)对斑马鱼胚胎发育的毒性作用,将斑马鱼胚胎暴露在0~50 ng/L的NGT下,对不同浓度NGT暴露下斑马鱼胚胎的基因表达情况进行比较分析,寻找不同浓度NGT影响下产生表达差异的基因,并通过差异基因富集分析对这些差异表达的基因进行功能注释,找到这些差异基因的功能和它们影响的代谢过程。结果表明:较低浓度NGT(5 ng/L)暴露下的斑马鱼胚胎,相比较高浓度NGT(50 ng/L)暴露下的斑马鱼胚胎,在基因表达层面表现出更强的差异性;低浓度NGT处理主要影响斑马鱼昼夜节律调控各类受体和转录因子的活性以及细胞的发育和成熟,高浓度NGT处理主要影响斑马鱼的视网膜代谢、细胞醛代谢、烯化合物代谢以及视黄酸类代谢等新陈代谢过程。To evaluate the toxic effects of norgestrel (NGT) on zebrafish embryonic development,the zebrafish embryos were exposed to 0-50 ng/L NGT separately.The comparative analysis of gene expression and functional notes between groups exposed to different concentrations of NGT was performed.As a result,zebrafish embryos exposed to 5 ng/L of NGT had a greater difference in gene expression than those exposed to 50 ng/L of NGT.Low concentrations of NGT mainly affect various receptors of circadian rhythm regulation and the activity of transcription factors and the development and maturation of cells in zebrafish.High concentrations of NGT mainly affect metabolic processes in zebrafish,including the retinal metabolism,cytoaldehyde metabolism,olefin compound metabolism and retinoic acid metabolism,etc.
