丙戊酸钠与多西环素治疗脊髓炎大鼠的行为学评分及对小胶质细胞、单核细胞及中性粒细胞水平的差异比较分析

摘  要:目的分析丙戊酸钠与多西环素治疗脊髓炎大鼠的行为学评分及对小胶质细胞、单核细胞及中性粒细胞水平的差异。方法构建致敏抗原诱导敏感实验动物诱发T细胞介导的反应性脊髓炎模型,并将大鼠分为模型组、丙戊酸钠组、多西环素组及空白对照组;模型组及空白对照组采用0.9%氯化钠注射液干预,丙戊酸钠组及多西环素组分别采用丙戊酸钠和多西环素干预;在免疫当天及药物干预结束后测量动物行为学变化情况,在干预结束后将大鼠处死取出脊髓,观察小胶质细胞情况;并在治疗结束后采集血液样品检测外周血中单核细胞及中性粒细胞水平。结果治疗后丙戊酸钠组及多西环素组大鼠行为学评分明显低于模型组,且差异存统计学意义(P<0.05),但丙戊酸钠组及多西环素大鼠行为学评分无明显差异(P<0.05);治疗后丙戊酸钠组及多西环素组大鼠胶质细胞水平显著低于模型组(P<0.05),但丙戊酸钠组与多西环素组无明显差异(P>0.05);治疗后丙戊酸钠组及多西环素组大鼠单核细胞及中性粒细胞水平显著低于模型组(P<0.05),但丙戊酸钠组与多西环素组无明显差异(P>0.05);大鼠体内小胶质细胞、单核细胞及中性粒细胞水平是影响大鼠行为学评分的独立性影响因素(P<0.05)。结论丙戊酸钠与多西环素治疗脊髓炎大鼠后可有效降低大鼠行为学评分,并显著改善小胶质细胞、单核细胞及中性粒细胞水平,但两种药物比较对各指标的影响无明显差异。Objective To analyze the behavioral scores of rats with myelitis treated with sodium valproate and doxycycline and the differences in the levels of microglia,monocytes and neutrophils.Methods The model of T-cell-mediated reactive myelitis induced by sensitized antigen in sensitive experimental animals was established,and the rats were divided into model group,sodium valproate group,doxycycline group and blank control group.The model group and blank control group were intervened by saline,while the sodium valproate group and doxycycline group were intervened by sodium valproate and doxycycline respectively.The changes of animal behavior were measured on the day of immunization and after drug intervention.After the intervention,rats were executed and the spinal cord was removed to observe the microglia.Blood samples were collected to detect the levels of peripheral mononuclear cells and neutrophils after the treatment.Results After treatment,the behavioral score of rats in sodium valproate group and doxycycline group was significantly lower than that in model group,and the difference was statistically significant(P<0.05),but there was no significant difference in behavioral score between sodium valproate group and doxycycline group(P<0.05);after treatment,the glial cell level of rats in sodium valproate group and doxycycline group was significantly lower than that in model group(P<0.05).Group A(P<0.05),but there was no significant difference between sodium valproate group and doxycycline group(P>0.05);after treatment,the levels of monocytes and neutrophils in sodium valproate group and doxycycline group were significantly lower than those in model group(P<0.05),but there was no significant difference between sodium valproate group and doxycycline group(P>0.05).The levels of microglia,monocytes and neutrophils in rats were independent factors affecting the behavioral score of rats(P<0.05).Conclusions Sodium valproate and doxycycline can effectively reduce the behavioral score of rats with myelitis,and significantly
