摘 要:目的探讨乳腺癌细胞中miR-769-5p的表达变化及其过表达对乳腺癌细胞增殖、凋亡、侵袭能力、迁移能力的影响及机制。方法通过qRT-PCR法检测乳腺癌细胞MCF-7、MDA-MB-231、SK-BR-3、HCC1806和正常乳腺上皮细胞HBL-100中miR-769-5p的表达。通过脂质体转染技术对HCC1806转染miR-769-5p模拟物(过表达组)和模拟物阴性对照(对照组),通过CCK-8实验检测细胞的增殖活性,通过流式细胞术检测细胞的凋亡率,通过Transwell实验检测细胞的迁移和侵袭能力,通过Western blotting法检测细胞中的核仁纺缍体相关蛋白1(NUSAP1)蛋白。双荧光素酶实验验证miR-769-5p对NUSAP1的靶向调控关系。结果miR-769-5p在乳腺癌细胞MCF-7、MDA-MB-231、SK-BR-3、HCC1806中的表达明显低于正常乳腺上皮细胞HBL-100(P均<0.05)。过表达组HCC1806细胞中miR-769-5p表达高于对照组,细胞增殖活性、侵袭和迁移能力低于对照组,凋亡率高于对照组,NUSAP1蛋白表达低于对照组(P均<0.05)。双荧光素酶实验显示miR-769-5p能靶向特异性结合NUSAP1的3′UTR区。结论miR-769-5p在乳腺癌细胞中呈低表达;体外过表达miR-769-5p能够抑制乳腺癌细胞的增殖、迁移和侵袭,促进其凋亡,其机制可能与miR-769-5p对NUSAP1基因的靶向调控有关。Objective To investigate the expression changes of miR-769-5p in breast cancer cells and the effects of overexpression of miR-769-5p on proliferation,apoptosis,invasion,and migration of breast cancer cells and their mechanism.Methods The expression of miR-769-5p in the breast cancer cells MCF-7,MDA-MB-231,SK-BR-3,HCC1806 and HBL-100 of normal breast epithelial cells was detected by qRT-PCR.HCC1806 cells were transfected with miR-769-5p mimics(overexpression group)and mimics negative control(control group)by liposome transfection technology.The proliferation activity of cells was detected by CCK-8,the apoptosis rate was detected by flow cytometry,the migration and invasion of cells were detected by Transwell experiment,and the nucleolar spindle-associated protein 1(NUSAP1)protein was detected by Western blotting.Double luciferase experiment verified the targeted regulation relationship of miR-769-5p with NUSAP1.Results The expression of miR-769-5p in the breast cancer cells MCF-7,MDA-MB-231,SK-BR-3 and HCC1806 was significantly lower than that in the normal breast epithelial cells(all P<0.05).The expression of miR-769-5p in HCC1806 cells in the overexpression group was higher than that in the control group,the cell proliferation activity,invasion and migration ability were lower than those in the control group,the apoptosis rate was higher than that in the control group,and the expression of NUSAP1 protein was lower than that in the control group(all P<0.05).Double luciferase assay showed that miR-769-5p could specifically bind to the 3′UTR region of NUSAP1.Conclusions MiR-769-5p expression is low in breast cancer cells.Overexpression of miR-769-5p in vitro can inhibit the proliferation,migration and invasion of breast cancer cells and promote its apoptosis.The mechanism may be related to the targeted regulation of miR-769-5p on NUSAP1 gene.
