卵巢癌组织中DR-NM23的表达及对细胞增殖和转移的影响

摘　　要：目的探究DR-NM23在卵巢癌组织中的表达及对细胞增殖和转移的影响。方法收集卵巢癌肿瘤组织和癌旁组织各68例,通过免疫组化染色检测DR-NM23表达水平,分析绝经、肿瘤大小、分化程度、局部浸润、淋巴转移和远端转移与DR-NM23表达水平的相关性。在SKOV3细胞系中过表达、沉默DR-NM23,分别得到DR-NM23组和SiDRNM23组,与对照组(NC组)相比,评估各组增殖、迁移和侵袭能力,以评估DR-NM23在卵巢癌组织中的作用机制。结果卵巢癌组织中DR-NM23平均染色强度为2.68±0.75,低于癌旁组织7.05±2.05,差异具有统计学意义(P<0.05)。卵巢癌组织中DR-NM23表达水平与是否绝经、肿瘤大小以及局部浸润无相关性,差异均无统计学意义(χ^(2)=0.418,0.864,2.915,均P>0.05)。低分化、出现淋巴转移和远端转移的组织中DR-NM23表达水平相对较低,差异均具有统计学意义(χ^(2)=6.678,6.845,8.215,均P<0.05)。SiDR-NM23组细胞的CCK-8吸光度(1.16±0.18)、迁移距离百分比(88.58%±5.48%)和侵袭细胞数(207.26±13.52个)高于NC组(0.68±0.11,62.34%±2.15%,125.54±8.15个),而DR-NM23组(0.47±0.07,45.12%±3.76%,59.57±5.02个)均低于NC组,差异均具有统计学意义(均P<0.05)。结论DR-NM23在卵巢癌组织中低表达,可能具有抑制肿瘤细胞增殖、迁移和侵袭的作用。Objective To explore the expression of DR-NM23 in ovarian cancer and its effect on cell proliferation and metastasis.Methods 68 cases of ovarian cancer tissues and adjacent tissues were collected clinically,and the DR-NM23 expression level was detected by immunohistochemistry,and the correlation of DR-NM23 expression level with menopause,tumor size,differentiation,local invasion,lymph node metastasis and distal metastasis was analyzed.Overexpression and silencing of DR-NM23 in SKOV3 cell line were used to obtain DR-NM23 group and Si-NM23 group,respectively.Compared with the control group(NC group),the proliferation,migration and invasion ability of each group were evaluated for the mechanism of DR-NM23 in ovarian cancer tissue.Results The average staining intensity of DR-NM23 in ovarian cancer tissue was(2.68±0.75),which was lower than that in adjacent tissues(7.05±2.05)with stastically significant difference(P<0.05).Whether there was no correlation in menopause,tumor size and local invasion with the expression level of DR-NM23 in ovarian cancer tissue,the differences not were statistically significant(χ^(2)=0.418,0.864,2.915,all P>0.05).The expression level of DR-NM23 was lower in tissues with poor differentiation,lymph node metastasis and distant metastasis,the differences were statistically significant(χ^(2)=6.678,6.845,8.215,all P<0.05).The CCK-8 absorbance(1.16±0.18),migration distance percentage(88.58%±5.48%)and invasive cells(207.26±13.52)of siDR-NM23 group was higher than that of NC group(0.68±0.11,62.34%±2.15%,125.54±8.15),while DR-NM23 group(0.47±0.07,45.12%±3.76%,59.57±5.02)was lower than that of NC group,the differences were stastically significant(all P<0.05).Conclusion DR-NM23 was lowly expressed in ovarian cancer tissue,might has the effect of inhibiting the proliferation,migration and invasion of tumor cells.
