摘 要:目的采用星点设计-响应面法优化三叉苦总黄酮提取工艺,并考察抗氧化活性。方法在单因素考察结果的基础上,对三叉苦总黄酮提取工艺中乙醇体积分数(%)、液料比(ml/g)、加热回流提取时间(min)3个因素进行优化,得到较优的总黄酮提取工艺。通过测定三叉苦总黄酮DPPH和ABTS自由基清除能力评价其抗氧化活性。结果三叉苦总黄酮最佳提取工艺:乙醇体积分数90%,液料比10 ml/g,提取时间30 min,该条件下实际的提取率2.99%,与预测值3.25%偏差较小,清除DPPH和ABTS自由基能力较强。结论运用响应面设计得到的方法稳定可靠,可用于三叉苦总黄酮的提取。三叉苦有显著的抗氧化能力,为其进一步开发和应用提供一定依据,在功能食品和药品开发方面具有较好前景。Objective To optimize the extraction process of total flavonoids from Melicope pteleifolia by central composite design(CCD)and response surface methodology,and evaluate the antioxidant activity.Methods Based on single factor tests,the ethanol volume fraction(%),the liquid-solid ratio(ml/g)and extraction time(min)were optimized by CCD method.The antioxidant activity of extraction from the Melicope pteleifolia was evaluated by DPPH and ABTS assays.Results The optimal extraction parameters of total flavonoids from Melicope pteleifolia were as follows:the ethanol volume fraction was 90%,the ratio of liquid to material was 10 ml/g and the extraction time was 30 min.Under this optimal condition,the yield of total flavonoids was 2.99%,which had a small deviation from the predicted value of 3.25%.The total flavonoids had strong scavenging ability to DPPH free radical and ABTS.Conclusions The extraction method of the total flavonoids from Melicope pteleifolia is stable and efficient.The extract has significant antioxidant capacity that provides a basis for the further development and application in the field of functional foods and drugs.
