摘 要:目的考察^(60)Co-γ射线辐照灭菌对白芍中6种成分[儿茶素、没食子酸、芍药内酯苷、芍药苷、1,2,3,4,6-O-五没食子酰葡萄糖(β-PGG)和苯甲酰芍药苷]含量的影响。方法采用Tnertsil^(®)ODS-2(250 mm×4.6 mm,5μm)色谱柱,流动相为A-乙腈,B-0.05%磷酸水溶液系统,梯度洗脱,检测波长230 nm,流速为1.0 ml/min,柱温30℃。分别选择剂量为0,2,5,8 kGy对白芍药材进行辐照,比较辐照前后活性成分含量的变化,并进行配对t检验考察其显著情况。结果白芍药材中儿茶素、没食子酸、芍药内酯苷、芍药苷、β-PGG和苯甲酰芍药苷分别在0.005~0.128,0.007~0.178,0.034~0.850,0.178~4.440,0.023~0.580,0.005~0.120 mg/ml范围内呈良好线性,平均加样回收率分别为99.1%,98.4%,99.9%,100.0%,99.8%和96.4%;辐照剂量不超过5 kGy时,各组分含量无显著变化(P>0.05)。结论为保证白芍药材安全有效,采用^(60)Co-γ射线对白芍药材进行辐照灭菌时,辐照剂量不宜超过5 kGy。Objective To study the effect of ^(60)Co-γ ray irradiation on the contents of catechin,gallic acid,albiflorin,paeoniflorin,1,2,3,4,6-penta-O-galloy-β-D-glucose and benzoylpaeoniflorin in Paeonia lactiflora Pall.Methods An Agilent Tnertsil^(®)ODS-2 column(250 mm×4.6 mm,5μm)was adopted and the wavelength of UV detection was 230 nm with the flow rate of 1.0 ml/min.The mobile phase consisted of 0.05%phosphoric acid(B)and acetonitrile(A)with a gradient elution,and the column temperature was 30℃.The irradiation of Paeonia lactiflora Pall.was done by choosing 0,2,5,8 kGy,the contents of the active ingredients were compared before and after irradiation,and a t-test was used to investigate its remarkable situation.Results The linear ranges of catechin,gallic acid,albiflorin,paeoniflorin,1,2,3,4,6-penta-O-galloy-β-D-glucose and benzoylpaeoniflorin in Paeonia lactiflora Pall.were 0.005-0.128 mg/ml,0.007-0.178 mg/ml,0.034-0.850 mg/ml,0.178-4.440 mg/ml,0.023-0.580 mg/ml and 0.005-0.120 mg/ml,respectively.The average recoveries were 99.1%,98.4%,99.9%,100.0%,99.8%and 96.4%,respectively.When the radiation dose was not more than 5 kGy,the contents of six components did not change significantly(P>0.05).Conclusion When the radiation dose of ^(60)Co-γ is not more than 5 kGy,the method of sterilization is safe and effective for Paeonia lactiflora Pall.
