摘 要:目的采用单细胞测序分析溃疡性结肠炎(ulcerative colitis, UC)患者肠道组织中巨噬细胞差异基因及其功能。方法挖掘数据集GSE150115,分析5例UC患者肠黏膜组织10×Genomics单细胞转录组测序数据,采用Seurat软件进行细胞亚群分类、巨噬细胞识别、细胞间差异基因筛选、GO和KEGG生物信息学分析。结果从10个细胞亚群中识别出巨噬细胞,筛选出细胞间差异基因,如CR1、BCL11A等。GO及KEGG分析发现,巨噬细胞的核糖体信号通路可能参与UC疾病过程。结论肠道巨噬细胞可能通过核糖体信号通路影响UC。Objective To analyze the differential expression genes and the function of macrophages in the intestine of patients with ulcerative colitis(UC). Methods Mining data set GSE150115, analyzing 10×Genomics single-cell transcriptome sequencing data of intestinal mucosal tissues of 5 UC patients, using Seurat software for cell subgroup classification, macrophage identification, genetic screening of differences among cells, GO and KEGG bioinformatics analysis.Results Macrophages were identified from 10 cell subgroups, and the differential genes among cells were screened out, such as CR1, BCL11 A, etc. GO and KEGG analysis found that the ribosome signaling pathway of macrophages were mainly involved in UC.Conclusion Intestinal macrophages may affect UC through ribosomal signaling pathways.
