摘 要:目的探讨竹荪多糖对砷中毒大鼠肝脏IL-6和TNF-α水平的影响。方法SPF级6周龄SD大鼠30只,雌雄各半,随机分为对照组(喂食正常饲料)、砷中毒组(喂食含砷50 mg/kg的饲料)及竹荪多糖组(喂食含砷50 mg/kg的饲料5个月后,以10 g/L竹荪多糖连续灌胃30 d);染毒终点记录大鼠体质量后麻醉处死、经心脏取血分离血清检测草转氨酶(AST)和谷丙转氨酶(ALT)水平,取大鼠肝脏计算脏器系数,取肝组织HE染色观察肝脏损伤情况,免疫组织化学法检测大鼠肝组织白细胞介素6(IL-6)、肿瘤坏死因子α(TNF-α)表达。结果与对照组相比,砷中毒组大鼠体质量明显下降,肝脏脏器系数升高,血清中ALT和AST的含量均升高,差异有统计学意义(P<0.05);与砷中毒组比较,竹荪多糖组大鼠体质量升高、肝脏脏器系数下降,血清中ALT和AST表达均有所下降,除ALT外,差异均有统计学意义(P<0.05);HE染色结果显示,砷中毒组大鼠肝脏有损伤,竹荪多糖组大鼠肝脏损伤程度有一定缓解;砷中毒组大鼠肝组织IL-6和TNF-α蛋白含量升高,竹荪多糖组大鼠肝组织中两指标下降(P<0.05)。结论竹荪多糖可能通过抑制炎症因子IL-6和TNF-α的表达,减轻砷暴露大鼠肝组织损伤。Objective To explore the antagonistic effect of polysaccharides from Dictyophora dictyophora against liver inflammation in arsenic poisoning rats,and to provide experimental basis for the development of the medicinal value of polysaccharides and the prevention and treatment of arsenic.Methods SPF grade 6-week-old SD rats(30),male and female,were randomly divided into control group(normal diet),arsenic group(arsenic 50 mg/kg diet),Dictyophora polysaccharide group(arsenic 50 mg/kg diet for 5 months,10 g/L Dictyophora polysaccharide every day for 30 days).At the end point of exposure,the body weight of each rat was recorded.Blood samples were collected from the heart to separate serum,and the expression of AST and ALT in serum was detected by biochemical analyzer.HE staining was used to detect liver injury,and the expressions of inflammatory factors interleukin-6(IL-6)and tumor necrosis factorα(TNF-α)in liver tissue were detected by immunohistochemistry.Results Compared with the control group,arsenic poisoning significantly decreased body weight,increased liver organ coefficient,and increased serum ALT and AST contents,with statistical significance(P<0.05);After Dictyophora polysaccharide intervention,the body weight of arsenic poisoning rats increased,the liver organ coefficient decreased,and the expression of ALT and AST in serum decreased,except ALT level,the differences were statistically significant(P<0.05);HE staining showed that arsenic exposure could induce liver damage in rats,and Dictyophora polysaccharide intervention had a certain alleviating effect.The levels of IL-6 and TNF-αin the liver tissue of arsenic-induced rats were increased,and the levels of TNF-αin the liver tissue of arsenic-induced rats were significantly decreased after the treatment with Dictyophora polysaccharide,and the TNF-αexpression was statistically significant(P<0.05).Conclusion Dictyopterus Polysaccharide juvola may inhibit the expression of inflammatory factors IL-6 and TNF-α,and alleviate the damage of liver tissue in rats
