摘  要:目的探讨趋化因子CCL18与乳腺癌临床病理特征的关系及其对乳腺癌细胞EMT过程的影响。方法选取行手术切除治疗的50例乳腺癌患者为研究对象,其中Ⅰ期3例,Ⅱ~Ⅲ期28例,Ⅳ期19例。采用免疫组织化学法检测乳腺组织中CCL18表达情况,分析其与临床病理特征的关系。选取乳腺癌细胞系BT-474和MDA-MB-231,分别用低浓度和高浓度CCL18处理48 h, Transwell侵袭实验检测其侵袭能力,Western Blot检测细胞E-cadherin和Vimentin蛋白表达情况。q-RT-PCR检测EMT相关转录因子的表达。结果 CCL18只在乳腺癌组织中表达,在癌旁组织中不表达(χ^(2)=47.059,P<0.01)。CCL18表达与乳腺癌患者TNM分期相关(χ^(2)=7.307,P<0.05),与患者年龄、绝经情况无关(χ^(2)=1.367,P>0.05;χ^(2)=1.937,P>0.05)。CCL18处理乳腺癌细胞后,BT-474和MDA-MB-231细胞的侵袭能力均显著增强(F=21.23,P<0.05;F=54.81,P<0.01):同时E-cadherin表达量降低(F=31.83,P<0.05;F=73.75,P<0.01),而Vimentin蛋白表达量升高(F=9.629,P<0.05;F=23.62,P<0.05)。上皮间充质转化(EMT)相关转录因子中,SNAIL因子被激活,表达量显著升高(F=21.57,P<0.05;F=24.36,P<0.05)。结论趋化因子CCL18的表达与乳腺癌临床病理特征相关,且CCL18通过激活SNAIL转录因子,促进乳腺癌细胞发生EMT,增强侵袭性。Objective To investigate the association of chemokine CCL18 with clinicopathological characteristics of breast cancer and its effect on EMT process in breast cancer cells.Methods 50 breast cancer patients who underwent breast cancer surgery were collected for the study, including 3 patients with stage Ⅰ,28 patients with stage Ⅱ~Ⅲ,and 19 patients with stage Ⅳ.CCL18 expression in breast tissue was assessed by immunohistochemistry and analyzed its relationship with different clinicopathological characteristics.The breast cancer cell lines BT-474 and MDA-MB-231 were treated with low and high CCL18 for 48 h.Transwell invasion assay was used to detect the invasive ability, Western Blot was used to detect the expression of E-cadherin and vimentin, q-RT-PCR was used to detect the expression of EMT related transcription factors.Results CCL18 was only expressed in breast cancer tissues and not in adjacent tissues(χ^(2)=47.059,P<0.01).There is no significant difference in the expression of CCL18 between patients over 50 and under 50,patients postmenopausal and premenopausal(χ^(2)=1.367,P>0.05;χ^(2)=1.937,P>0.05).The expression of CCL18 in patients with stage Ⅲ to Ⅳ was significantly higher than that in patients with stage Ⅰ to Ⅱ(χ^(2)=7.307,P<0.05).CCL18 enhanced the invasive ability of both BT-474 and MDA-MB-231 cells(F=21.23,P<0.05;F=54.81,P<0.01),meanwhile, expression of E-cadherin was decreased(F= 31.83,P<0.05;F=73.75,P<0.01),expression of vimentin protein was increased(F=9.629,P<0.05;F=23.62,P<0.05).Among epithelial mesenchymal transition(EMT)-related transcription factors, snail was activated(F=21.57,P<0.05;F= 24.36,P<0.05).Conclusions The expression of chemokine CCL18 is associated with breast cancer clinicopathological features, and CCL18 activates EMT of breast cancer cells by snail transcription factor.
