摘  要:研究旨在制作一种可以通过单胃动物胃液的酸性环境并在肠液中释放的姬松茸多糖-嗜酸乳杆菌合生元。试验采用微胶囊法,以海藻酸钠和姬松茸多糖作为复合壁材包裹嗜酸乳杆菌,使用壳聚糖进行2次包被,测定微胶囊的包埋率、在人工胃液中的保护性及在人工肠液中的释放性,评估微胶囊的效果;使用牛津杯法测定合生元的抑菌效果。结果显示,当海藻酸钠浓度为1.0%,姬松茸多糖浓度1.5%,固化时间为4 min时,制得的微胶囊具有最好的包埋率,极显著高于对照组(P<0.01)。经胃液作用2 h后,合生元微胶囊活菌数仍有4.07×10^(7)CFU/g。3组微胶囊在模拟肠液中释放最大活菌数均在2 h处,各组间差异极显著(P<0.01)。ASL1的抑菌性优于嗜酸乳杆菌溶液(LA)、姬松茸多糖溶液(AB)。研究表明,合生元微胶囊对嗜酸乳杆菌具有较好的包埋效果和保护作用,可显著提高抑菌活性。The study was to prepare a Agaricus blazei polysaccharide Lactobacillus acidophilus synbiotic that could pass through the acidic environment of gastric juice and be released in intestinal juice. Sodium alginate and Agaricus blazei polysaccharide were used as composite wall materials to wrap Lactobacillus acidophilus, and chitosan was used for secondary coating. The encapsulation rate, protection in artificial gastric juice and release in artificial intestinal juice were measured to evaluate the effect of microcapsules. Oxford cup was used to determine the bacteriostatic effect of synbiotics.The results showed that when the sodium alginate concentration was 1.0%, the Agaricus blazei polysaccharide concentration was 1.5%, the curing time was 4 min, and the prepared microcapsules had the best encapsulation rate,extremely higher than control group(P<0.01). After being exposed to gastric juice for 2 h, the viable count of synbiotic microcapsules was still 4.07×10;CFU/g. The maximum number of viable bacteria released by the three groups of microcapsules in the simulated intestinal fluid was at 2 h, and the differences among the groups were extreme(P<0.01).The antibacterial activity of ASL1 was better than that of Lactobacillus acidophilus solution(LA) and Agaricus blazei polysaccharide solution(AB). The experiment indicates that synbiotic microcapsules have better embedding effect and protective effect on Lactobacillus acidophilus, which can significantly improve the antibacterial activity.
