摘　　要：目的探讨去整合素金属蛋白酶12(ADAM12)在结直肠癌中的表达水平及其与结直肠癌患者临床病理特征之间的关系。方法收集安徽医科大学附属巢湖医院胃肠外科2020年4月-2021年2月行手术治疗的83例结直肠癌患者的癌组织及对应的癌旁组织标本,采用免疫组化的方法检测结直肠癌及癌旁组织中ADAM12蛋白的表达量,分析ADAM12在结直肠癌中表达水平及其与患者临床病理特征的关系。结果结直肠癌患者中ADAM12高表达占比高于低表达患者,且ADAM12在癌组织中表达高于癌旁组织,差异有统计学意义(P<0.05);ADAM12主要在结直肠癌的胞浆和细胞膜中表达,呈棕褐色或棕黄色,在正常组织中呈低表达或不表达;不同年龄、性别、肿瘤位置、肿瘤大小、分化程度、脉管侵犯以及神经侵犯的患者ADAM12表达比较,差异无统计学意义(P>0.05);不同肿瘤浸润深度、淋巴结转移、TNM分期的患者ADAM12表达比较,差异有统计学意义(P<0.05)。结论 ADAM12在结直肠癌患者中呈高表达,高表达的ADAM12在结直肠癌发生发展过程中可能起到促癌的作用并与结直肠癌的侵袭转移密切相关,且可能是结直肠癌靶向治疗的一个潜在分子标志物。Objective To investigate the expression level of a disintegrin and metalloprotease 12(ADAM-12) in colorectal cancer and its relationship with clinicopathological characteristics of colorectal cancer patients.Methods The cancer tissues and corresponding adjacent tissues of 83 patients with colorectal cancer who underwent surgical treatment in the Department of Gastrointestinal Surgery of Chaohu Hospital of Anhui Medical University from April 2020 to February 2021 were collected.The expression of ADAM12 protein in colorectal cancer and adjacent tissues was detected by immunohistochemistry.The expression level of ADAM12 in colorectal cancer and its relationship with clinicopathological features were analyzed.Results The proportion of ADAM12 high expression in colorectal cancer patients was higher than that in patients with low expression,and the expression of ADAM12 in cancer tissues was higher than that in adjacent tissues,the differences were statistically significant( P<0.05).ADAM12 was mainly expressed in cytoplasm and cell membrane of colorectal cancer,brown or brown-yellow,and low or no expression in normal tissues.There was no significant difference in ADAM12 expression between patients with different age,gender,tumor location,tumor size,differentiation degree,vascular invasion and nerve invasion(P>0.05).The expression of ADAM12 in patients with different tumor invasion depth,lymph node metastasis and TNM stage was statistically significant(P<0.05).Conclusion ADAM12 is highly expressed in patients with colorectal cancer.High expression of ADAM12 may play a role in promoting cancer development and is closely related to the invasion and metastasis of colorectal cancer,and may be a potential molecular marker for targeted therapy of colorectal cancer.
