摘  要:目的观察山仙颗粒对H22肝癌移植瘤的抑制作用及肿瘤组织中干扰素-γ(IFN-γ)、白细胞介素-6(IL-6)、转化生长因子-β(TGF-β)含量的影响,探讨山仙颗粒对H22肝癌移植瘤微环境中淋巴细胞活性的调控作用。方法取50只雄性昆明小鼠,随机选择40只小鼠建立H22肝癌移植瘤模型,其余10只小鼠作为空白组。造模24 h后将小鼠随机分为模型组及山仙颗粒低、中、高剂量组各10只,山仙颗粒低、中、高剂量组分别给予山仙颗粒0.025 g/mL、0.05 g/mL、0.1 g/mL灌胃,空白组和模型组给予等量生理盐水灌胃,均早晚各1次,连续30 d。颈椎脱臼法处死小鼠,剥离瘤组织,计算抑瘤率;ELISA法检测H22移植瘤组织中IFN-γ、IL-6、TGF-β含量。结果山仙颗粒低、中、高剂量组H22肝癌移植瘤的抑瘤率随用药剂量增加而增高,各组间比较差异均有统计学意义(P均<0.05)。与空白组比较,模型组移植瘤组织中IFN-γ含量明显降低(P<0.05),IL-6、TGF-β含量均明显增高(P均<0.05);与模型组比较,山仙颗粒各组移植瘤组织中IFN-γ含量明显增高(P均<0.05),IL-6、TGF-β含量均明显降低(P均<0.05);山仙颗粒高剂量组移植瘤组织中IFN-γ、IL-6、TGF-β含量变化更明显,其次为山仙颗粒中剂量组,各组间比较差异均有统计学意义(P均<0.05)。结论山仙颗粒具有抑制小鼠H22肝癌移植瘤生长的作用,其机制可能与调控肿瘤微环境中淋巴细胞活性,影响IFN-γ、IL-6、TGF-β的表达,从而提高机体抗肿瘤免疫功能相关。Objective It is to observe the inhibitory effect of Shanxian granule on H22 liver cancer transplanted tumor and its effect on the contents of interferon-γ(IFN-γ), interleukin-6(IL-6) and transforming growth factor-β(TGF-β) in tumor tissue, to explore the regulatory effect of Shanxian granules on the activity of lymphocytes in the microenvironment of H22 hepatocellular carcinoma xenografts. Methods Fifty male Kunming mice were selected, in which 40 mice were randomly selected to establish the H22 liver cancer xenograft models, and the remaining 10 mice were used as the blank group. 24 hours after modeling, the mice were randomly divided into model group and Shanxian granule low-dose, medium-dose, and high-dose groups, 10 mice in each group. Shanxian granule low-dose, middle-dose and high-dose groups were respectively given Shanxian granules 0.025 g/mL, 0.05 g/mL, 0.1 g/mL by gavage, and the blank group and model group were given the same amount of normal saline by gavage, once in the morning and once in the evening, all treated for 30 days. The mice were killed by cervical dislocation, the tumor tissue was stripped, and the tumor inhibition rate was calculated. The contents of IFN-γ, IL-6 and TGF-β in H22 transplanted tumor tissue were detected by ELISA. Results The tumor inhibition rate of H22 hepatocellular carcinoma xenografts in the low, medium and high dose groups of Shanxian granules were increased with the increase of the drug dose, and there were significant differences among the groups(P all<0.05). Compared with the blank group, the content of IFN-γ in the transplanted tumor tissue of the model group was significantly decreased(P<0.05), while the contents of IL-6 and TGF-β were significantly increased(all P<0.05). Compared with the model group, the content of IFN-γ in the transplanted tumor tissue of each Shanxian granule group was significantly increased(P all<0.05), while the contents of IL-6 and TGF-β were significantly decreased(P all<0.05).
