摘　　要：目的探讨硫酸乙酰肝素蛋白多糖在唾液腺腺样囊性癌神经侵袭过程中的表达及作用。方法通过Real time PCR技术检测硫酸乙酰肝素蛋白多糖(GPC1、GPC2、GPC3、GPC4、GPC5、GPC6、HSPG2、SDC1、SDC2、SDC3、SDC4)在腺样囊性癌细胞SACC-83细胞与人雪旺细胞中的表达。免疫组织化学染色,检测SDC1、SDC4在腺样囊性癌伴神经侵袭肿瘤组织中的表达。结果Real time PCR结果显示,不同的硫酸乙酰肝素蛋白多糖在SACC-83细胞呈现不同的表达水平,SDC1、SDC4在SACC-83细胞中相对表达量分别是人雪旺细胞的5.49倍和2.09倍;SDC1与SDC4在腺样囊性癌肿瘤组织中呈阳性表达,其中SDC4在伴有周围神经侵袭肿瘤组织中表达高于远离神经侵袭部位表达(P<0.05)。结论硫酸乙酰肝素蛋白多糖SDC4可能参与了唾液腺腺样囊性癌的周围神经侵袭过程。Objective To analyze the heparan sulfate proteoglycans expression in salivary adenoid cystic carcinoma with perineural invasion.Methods The Real time PCR was used to detect the mRNA expression of heparan sulfate proteoglycans(GPC1、GPC2、GPC3、GPC4、GPC5、GPC6、HSPG2、SDC1、SDC2、SDC3、SDC4)in salivary adenoid cystic carcinoma cell line(SACC-83)and human schwann cells.Immunohistochemical staining was used to detect SDC1 and SDC4 expression in salivary adenoid cystic carcinoma with perineural invasion.Results HSPGs were expressed at different levels in the SACC-83 cell line by relative-quantitative Real time PCR detection.Compared with human schwann cells,the mRNA expression of SDC1 and SDC4 in SACC-83 cells increased as 5.49-fold and 2.09-fold respectively.SDC1 and SDC4 showed positive expression in salivary adenoid cystic carcinoma,SDC4 expressed particularly at higher level in tumor with perineural invasion compared with that away from the perineural invasion(P<0.05).Conclusion SDC4 may involve in the perineural invasion process of salivary adenoid cystic carcinoma.
