摘 要:目的研究4 Gy X线照射对骨肉瘤细胞迁移能力的影响及其机制。方法将143B细胞和U2OS细胞分为对照组和4 Gy照射组,分别于照射后12、24、36和48 h,采用Transwell法检测细胞的迁移能力,Western印迹检测细胞MMP-2和MMP-9蛋白表达水平;siRNA法干扰细胞MMP-2/MMP-9表达。结果与对照组相比,4 Gy X线照射后,143B细胞和U2OS细胞12和48 h的迁移能力未发生显著变化(P>0.05),24和36 h的迁移能力均显著增强(P<0.05);同时,两种细胞的MMP-2和MMP-9蛋白表达水平在12和48 h无显著变化(P>0.05),在24及36 h显著升高(P<0.05);干扰143B细胞和U2OS细胞中的MMP-2/MMP-9表达后,两种细胞的MMP-2/MMP-9蛋白表达水平及迁移能力均明显降低(P<0.05)。结论4 Gy X线照射可通过上调MMP-2/MMP-9表达水平增强143B细胞和U2OS细胞的迁移能力。Objective To investigate the effect of 4 Gy X-ray irradiation on migration of osteosarcoma cells and the mechanism.Methods 143 B cells and U2 OS cells were divided into control and 4 Gy irradiation groups.The cell migration ability was detected using Transwell method at 12,24,36 and 48 h after irradiation,and the cell MMP-2 and MMP-9 protein expression levels were detected by Western Blot at 12,24,36 and 48 h after irradiation.siRNA was used to interfere with cellular MMP-2/MMP-9 expressions.Results Compared with the control group,the migration ability of143 B cells and U2 OS cells did not change significantly(P>0.05)at 12 and 48 h,but was significantly enhanced(P<0.05)at 24 and 36 h after 4 Gy X-ray irradiation.Meanwhile,the expression levels of MMP-2 and MMP-9 proteins in either type of cell did not change significantly(P>0.05)at 12 and 48 h,and were significantly increased(P<0.05)at 24 and 36 h.At the same time,the MMP-2 and MMP-9 protein expression levels of both cells hardly changed at 12 and 48 h(P>0.05)but increased significantly at 24 and 36 h(P<0.05).After interference with MMP-2/MMP-9 expression in 143 B cells and U2 OS cells,the MMP-2/MMP-9 protein expression levels and migration ability of both types of cells decreased significantly(P<0.05).Conclusion 4 Gy X-ray irradiation can enhance the migration ability of 143 B cells and U2 OS cells by upregulating the expression level of MMP-2/MMP-9.
