摘 要:利用病毒宏基因组学(viral metagenomics)方法,本研究从95份林麝粪便样品中检测到1株新型圆环病毒样(Circoviruslike)病毒,经过拼接获得其全基因组序列,该毒株命名为ls-cyc。其基因组全长约3552 nt,GC含量39.00%,含有2个开放阅读框(ORF),即ORF1和ORF2。其中ORF1长度为1386 nt,ORF2长度为1953 nt。ORF1编码的氨基酸序列与来自猪血清圆环状病毒的(MH170058.1)序列相似度为34.31%。系统进化分析表明其与来自猪的圆环状病毒(MH170058.1)和鸭相关环状病毒(NC_030134.1)为同一进化分支。该病毒是否与林麝特定疾病相关有待于进一步流行病学研究。A novel circovirus-like virus was discovered from 95 fecal samples of musk deer using viral metagenomics method.The newly discovered virus was designated as ls-cyc.The complete genome sequence was acquired by sequence assembly,of which the genome length was 3552 nt and the GC content was 39.00%.Genomic structure analysis showed that the virus contained two open reading frames,namely ORF1 and ORF2.The ORF1 length was 1386 nt and ORF2 was 1953 nt.The amino acid sequence encoded by ORF1 showed 34.31%similarity to a pig serum circular circovirus(MH170058.1).Phylogenetic analysis based on the REP protein showed that ls-cyc shared the same evolutionary branch as the circular circoviruses from pigs(MH170058.1)and ducks(NC_030134.1).Further epidemiological studies were needed to determine whether the virus was associated with specifi c diseases of musk deer.
