c-IAP1和Caspase-3在喉癌及癌旁组织中的表达及临床意义

摘　　要：目的检测凋亡抑制蛋白c-IAP1及天冬氨酸特异性蛋白酶Caspase-3在喉癌及癌旁正常上皮组织中的表达及两种蛋白之间的相关性,从而进一步探讨两种蛋白在喉癌发生发展中的作用及临床意义。方法将60例喉癌组织和60例癌旁组织的石蜡组织切片进行免疫组织化学(IHC)染色,检测c-IAP1及Caspase-3蛋白的表达水平。结果c-IAP1蛋白在喉癌组织中的阳性表达率高于癌旁正常上皮组织(P<0.05)。Caspase-3蛋白在喉癌组织中的阳性表达率低于癌旁正常上皮组织(P<0.05)。喉癌组织中c-IAP1、Caspase-3蛋白的阳性表达率与吸烟、饮酒、分化程度、淋巴结转移及TNM分期相关(P<0.05),而与年龄、性别、临床分型等均无相关性(P>0.05)。结论c-IAP1蛋白在喉癌组织中的表达升高,Caspase-3蛋白在喉癌组织中的表达降低。在喉癌组织中c-IAP1与Caspase-3蛋白的表达呈负相关(r=-0.517,P<0.001)。上述两种蛋白的异常表达与吸烟、饮酒、分化程度、淋巴结转移及TNM分期相关,而与年龄、性别、临床分型等均无关。进一步推测c-IAP1、Caspase-3蛋白可能在喉癌的发生和发展过程中发挥了一定的作用。Objective To study the expression of apoptosis inhibitor protein C-IAP1 and aspartic acid specific protease caspase-3 in laryngeal cancer and adjacent normal epithelial tissues and the correlation between the two proteins were detected,so as to further explore the role of the two proteins in the occurrence and development of laryngeal cancer and its clinical significance.Methods Immunohistochemistry(IHC)staining was performed on paraffin sections of 60 laryngeal carcinoma tissues and 60 adjacent tissues to detect the expression levels of C-IAP1 and Caspase-3 proteins.Results The positive expression rate of C-IAP1 protein in laryngeal carcinoma tissues was higher than that in adjacent normal epithelial tissues(P<0.05);but the positive expression rate of Caspase-3 protein in laryngeal carcinoma tissues was lower than that in adjacent normal epithelial tissues(P<0.05);the positive expression rates of C-IAP1 and Caspase-3 protein in laryngeal carcinoma tissues were correlated with smoking,alcohol consumption,differentiation degree,lymph node metastasis and TNM stage(P<0.05),but had no correlation with age,gender,clinical classification(P>0.05).Conclusion The expression of C-IAP1 protein was increased and caspase-3 protein was decreased in laryngeal cancer tissues.There was a negative correlation between c-IAP1 and caspase-3 protein expression in laryngeal carcinoma(r=-0.517,P<0.001).The abnormal expression of these two proteins was correlated with smoking,alcohol consumption,differentiation degree,lymph node metastasis and TNM stage,but not with age,sex and clinical typing.It is further speculated that C-IAP1 and Caspase-3 proteins may play a certain role in the occurrence and development of laryngeal carcinoma.
