摘 要:目的:探讨青蒿琥酯(artesunate,ART)对人舌鳞癌CAL27细胞的抑制作用及可能的机制。方法:用不同剂量的ART预处理CAL27细胞,然后使用CCK-8和集落形成法进行细胞活力分析,流式细胞仪检测细胞凋亡,并通过划痕实验和Transwell小室测定细胞迁移和侵袭能力。通过qRT-PCR测定基质金属蛋白酶9(matrix metallopeptidase 9,MMP-9)和血管内皮细胞生长因子(vascular endothelial growth factor,VEGF)mRNA表达量,利用Western印迹法检测各浓度ART处理的CAL27细胞的MMP-9和VEGF的表达情况及信号传导与转录激活因子3(signal transducer and activator of transcription 3,STAT3)信号的活化情况。采用SPSS 19.0软件包对数据进行统计学分析。结果:青蒿琥酯以剂量依赖性方式显著抑制CAL27细胞的增殖(P<0.01)、侵袭(P<0.01)和迁移(P<0.01),诱导细胞凋亡(P<0.01)。ART不仅显著降低CAL27细胞中MMP-9和VEGF mRNA的表达,呈剂量依赖性(P<0.05),而且抑制p-STAT3、MMP-9和VEGF的蛋白表达。结论:青蒿琥酯可抑制人舌鳞癌细胞CAL27的增殖、迁移和侵袭,可能通过抑制STAT3信号通路而发挥抗肿瘤作用。PURPOSE:To investigate the anticancer effect of artesunate(ART)on human tongue squamous cell carcinoma(CAL27)cells and its possible mechanism.METHODS:CAL27 cells was pretreated with different doses of ART.Then,CCK-8 and colony forming methods were used for cell viability analysis,cell apoptosis was detected by flow cytometry,and cell migration and invasion capacity were determined by scratch test and Transwell chamber method.In addition,the expression of matrix metalloproteinase-9(MMP-9)and vascular endothelial growth factor(VEGF)mRNA was measured by qPCR,and Western blot was used to detect the expression of MMP-9 and VEGF and activation of STAT3 signal in CAL27 cells treated with ART at various concentrations.SPSS 19.0 software package was used for statistical analysis of the data.RESULTS:Artesunate significantly inhibited proliferation(P<0.01),invasion(P<0.01)and migration(P<0.01)of CAL27 cells,and induced apoptosis of CAL27 cells(P<0.01)in a dose-dependent manner.ART not only significantly reduced the expression of MMP-9 and VEGF mRNA in CAL27 cells in a dose-dependent manner(P<0.05),but also inhibited the protein expression of p-STAT3,MMP-9 and VEGF.CONCLUSIONS:Artesunate can inhibit the proliferation,migration and invasion of CAL27 cells,which may exert antitumor effects by inhibiting the STAT3 signaling pathway.
