摘 要:目的研究石杉碱甲对β-淀粉样蛋白间接诱导的神经元线粒体毒性的保护作用。方法采用transwell构建小胶质细胞和神经元细胞的共培养体系,实验利用共培养体系设置对照组、Aβ_(1-42)组、HupA组。通过检测活性氧(ROS)、总SOD活性和MDA水平、ATP和MTT水平来评估石杉碱甲在Aβ_(1-42)诱导的PC12细胞线粒体毒性过程中的保护作用。结果①石杉碱甲可以显著减少Aβ_(1-42)诱导的PC12细胞ROS、MDA水平;②石杉碱甲明显提高PC12细胞的T-SOD和ATP水平。结论石杉碱甲通过改善Aβ_(1-42)间接诱导的PC12细胞线粒体毒性发挥神经细胞活性的保护作用。Objective To investigate the protective effect of huperzine A(HupA)on mitochondrial toxicity induced by amyloid-β(Aβ).Methods A coculture system was constructed between microglia and neuronal cells through transwell,which was then used to set up the control,Aβ_(1-42),and HupA groups.The protective role of huperzine in the mitochondrial toxicity of PC12 cells induced by Aβ_(1-42) indirectly was assessed by examining reactive oxygen species(ROS),total SOD activity,and the levels of MDA,ATP,and MTT.Results①Huperzine A significantly reduced the ROS,MDA levels of PC12 cells induced by Aβ_(1-42).②Huperzine A significantly increased the T-SOD and ATP levels in PC12 cells.Conclusion Huperzine A exerts a protective role of nerve cell activity by alleviating mitochondrial toxicity induced by A protein in PC12 cells.
