摘 要:目的探寻miR-128在种植体周围炎骨破坏中的作用。方法通过Real-time PCR检测miR-128和TRAP在10例种植体周围炎患者炎性组织和10例健康人牙龈组织中的表达情况。将miR-128 inhibitor(抑制物)及mimics(模拟物)转染入破骨细胞后,通过CCK8检测miR-128对破骨细胞增殖能力的影响。通过Real-time PCR及Western blot检测转染miR-128 inhibitor后,TRAP的基因及蛋白在miR-128 inhibitor组、Mock组(不转染组)及inhibitor NC组(对照组)的表达情况;同时检测转染miR-128 mimics后,TRAP的基因及蛋白在miR-128 mimics组、Mock组(不转染组)及Negative control组(对照组)的表达情况。结果miR-128及TRAP在10例种植体周围炎患者炎性组织中的表达高于10例健康牙龈组织(P<0.05)。miR-128与TRAP存在中度相关性(r=0.527,P<0.05);CCK8结果显示,miR-128 inhibitor及mimics转染破骨细胞24、48、72 h后,miR-128 mimics促进破骨细胞增殖能力,miR-128 inhibitor抑制破骨细胞增殖能力(P<0.05);miR-128 inhibitor转染破骨细胞后,TRAP的基因及蛋白表达降低(P<0.05)。miR-128 mimics转染破骨细胞后,TRAP的基因及蛋白表达增高(P<0.05)。结论miR-128在种植体周围炎组织中表达增高,在种植体周围炎中起到了促进骨破坏的作用。Objective To explore the role of miR-128 in the destruction of inflammatory bone around implants.Methods Real-time PCR was used to detect the expression of miR-128 and tartrate-resistant acid phosphatase(TRAP)in inflammatory tissues of 10 patients with peri-implantitis and 10 healthy gingival tissues.After transfecting miR-128 inhibitor(inhibitor)and miR-128 mimics(mimetic)into osteoclasts,CCK8 was used to detect the effect of miR-128 on the proliferation of osteoclasts.Real-time PCR and Western blot were used to detect the expression of TRAP gene and protein in miR-128 inhibitor group,Mock group(non-transfection group)and Negative NC group(control group)after transfection of miR-128 inhibitor;After transfection of miR-128 mimics,the expressions of TRAP gene and protein in the miR-128 mimics group,Mock group(non-transfection group)and Negative control group(control group)were detected.Results The expression of miR-128 and TRAP in the inflammatory tissues of 10 patients with peri-implantitis was higher than that of 10 healthy gingival tissues(P<0.05).There is a moderate correlation between miR-128 and TRAP(r=0.527,P<0.05);CCK8 results showed that miR-128 inhibitor and mimics transfected osteoclasts for 24,48 and 72 h,miR-128 mimics promoted osteoclast proliferation ability,miR-128 inhibitor inhibits osteoclast proliferation ability(P<0.05);after miR-128 inhibitor transfects osteoclasts,TRAP gene and protein expression decreases(P<0.05).After miR-128 mimics was transfected into osteoclasts,TRAP gene and protein expression increased(P<0.05).Conclusion The expression of miR-128 is increased in peri-implant inflammation tissue,and plays a role in promoting bone destruction in peri-implantitis.
