摘  要:目的探讨放射性视神经病变(RION)相关microRNA的差异表达及其应用价值。方法应用直线加速器对视网膜神经节细胞(RGC-5)进行照射,吸收剂量分别为0、2、4和8 Gy,分别于照射前和照射后1~7 d在显微镜下观察细胞形态的变化,测定细胞的存活状态,Real-time PCR检测microRNA在RGC-5细胞照射前后的表达情况。同时收集接受头颈部放疗并发生RION患者16例(RION组)和未发生RION患者16例(对照组),比较放疗前后外周血中microRNA表达情况。结果照射后4 Gy组和8 Gy组较其他各组细胞生长和形态发生明显改变;随着照射时间的增加,2、4、和8 Gy组细胞存活率较0 Gy组均下降(均P<0.05)。RGC-5细胞接受8 Gy照射7 d后,miR-192和miR-34a表达明显上调(均P<0.05);RION组放疗后miR-192和miR-34a均高于对照组(均P<0.05)。miR-192预测RION的敏感性和特异性分别为93.75%和81.25%,miR-34a预测RION的敏感性和特异性分别为87.75%和75.00%。结论 miR-192和miR-34a与RION有关,可作为预测其发生的血清学标志物。Objective To investigate the differential expression and application value of radiation induced optic neuropathy(RION) related microRNAs.Methods Retinal ganglion cells(RGC-5) were irradiated with linear accelerator at doses of 0,2,4 and 8 Gy,respectively.Morphological changes of RGC-5 cells were observed under a microscope before irradiation and one to seven days after irradiation,respectively,and survival status of the cells was determined.Real-time PCR was used to detect microRNA expression before and after irradiation of RGC-5 cells.Meanwhile,16 patients with head and neck radiotherapy and RION occurrence(RION group) and 16 patients without RION occurrence(control group) were collected to compare microRNA expression in peripheral blood before and after radiotherapy.Results The cell growth and morphology of 4 Gy group and 8 Gy group were significantly changed compared with other groups after irradiation.With the increase of irradiation time,the survival rate of cells in 2,4,and 8 Gy groups decreased compared with that in 0 Gy group(all P <0.05).After RGC-5 cells were irradiated with 8 Gy for seven days,the expressions of miR-192 and miR-34a were significantly up-regulated(all P<0.05).Both miR-192 and miR-34A in the RION group were higher than those in the control group after radiotherapy(all P <0.05).The sensitivity and specificity of miR-192 to predict RION were 93.75% and 81.25%,respectively,and miR-34a to predict RION were 87.75% and 75.00%,respectively.Conclusions miR-192 and miR-3 4 a are related to RION and can be used as serological markers to predict its occurrence.
