摘 要:目的探讨金线莲苷对人肝癌HepG2细胞增殖、迁移和侵袭的影响。方法选取不同浓度金线莲苷作用于HepG2细胞,用四唑盐比色法(MTT)和细胞增殖-毒性实验(CCK-8)检测HepG2细胞的增殖和活性,Transwell实验检测细胞的运动能力,划痕实验检测细胞的迁移能力。结果随着金线莲苷浓度的增加,HepG2细胞的增殖抑制率逐渐上升,存活率逐渐下降,迁移和侵袭的细胞数逐渐减少;而随着作用时间的增加,细胞划痕面积越来越小,迁移率越来越高。结论金线莲苷能明显抑制人肝癌HepG2细胞的增殖活性,对HepG2细胞的侵袭和迁移能力也有抑制作用,且存在剂量依赖性。Objective To investigate the effects of Kinsenoside on the proliferation,migration and invasion of human Hepatoma HepG2 cells.Methods HepG2 cells were treated with different concentrations of Kinsenoside.The proliferation and activity of HepG2 cells were detected by tetrazolium salt colorimetry(MTT)and cell proliferation-toxicity assay(CCK-8),the mobility of cells was detected by Transwell assay,and the migration ability of cells was detected by scratch assay.Results With the increase of Kinsenoside concentration,the proliferation inhibition rate of HepG2 cells gradually increased,the survival rate gradually decreased,and the number of migrating and invasive cells gradually decreased.With the increase of treatment time,the cell scratch area became smaller and the migration rate became higher.Conclusion Kinsenoside can significantly inhibit the proliferation activity of human hepatoma HepG2 cells,and also inhibit the invasion and migration ability of them in a dose-dependent manner.
