摘　　要：目的观察胃癌患者癌组织JADE家族PHD指因子1(jade family PHD finger 1, JADE1)及组蛋白乙酰转移酶结合起始复合物1(histone acetyltransferase binding to ORC1, HBO1)表达情况,探讨其与胃癌患者预后的关系。方法 92例胃癌患者均行手术切除治疗,取手术切除的胃癌组织和癌旁组织,采用免疫组织化学法检测2种组织JADE1、HBO1阳性表达率;比较不同临床病理特征的胃癌患者癌组织JADE1、HBO1阳性表达率,采用Spearman相关性分析胃癌组织JADE1与HBO1表达的相关性。采用Kaplan-Meier法绘制生存曲线,比较JADE1、HBO1阳性表达者与阴性表达者3年总体生存率;采用多因素Cox回归分析胃癌患者预后不良的影响因素。结果 JADE1阳性染色主要位于细胞质和细胞膜,HBO1阳性染色主要位于细胞核。胃癌组织JADE1、HBO1阳性表达率(67.4%、72.8%)均高于癌旁组织(13.0%、10.9%)(χ^(2)=56.511,P<0.001;χ^(2)=72.559,P<0.001)。胃癌组织中JADE1与HBO1表达呈正相关(r=0.491,P<0.001)。临床分期Ⅲ期的胃癌患者癌组织JADE1、HBO1阳性表达率(88.9%、92.6%)均高于临床分期Ⅰ~Ⅱ期者(58.5%、64.6%)(χ^(2)=8.037,P=0.005;χ^(2)=7.545,P=0.006),有淋巴结转移者癌组织JADE1、HBO1阳性表达率(87.9%、90.9%)高于无淋巴结转者(55.9%、62.7%)(χ^(2)=9.829,P=0.002;χ^(2)=8.503,P=0.004)。JADE1阳性表达者3年总体生存率(60.7%)低于阴性表达者(83.3%)(χ^(2)=17.562,P<0.001),HBO1阳性表达者3年总体生存率(60.6%)低于阴性表达者(88.0%)(χ^(2)=18.821,P<0.001)。胃癌组织JADE1阳性表达(HR=2.141,95%CI:1.293~3.663,P=0.004)、HBO1阳性表达(HR=3.560,95%CI:1.921~6.379,P<0.001)、临床分期Ⅲ期(HR=2.730,95%CI:1.442~5.199,P=0.003)及有淋巴结转移(HR=5.236,95%CI:1.268~8.266,P=0.025)是胃癌患者预后不良的独立危险因素。结论胃癌患者癌组织JADE1、HBO1阳性表达率升高,二者表达呈正相关,胃癌组织JADE1、HBO1阳性表达提示胃癌患者可能预后不良。Objective To observe the expressions of Jade family PHD finger 1(JADE1) and histone acetyltransferase binding to ORC1(HBO1) in gastric cancer tissues, and to investigate their relationships with the prognosis of patients with gastric cancer. Methods Ninety-two patients with gastric cancer underwent surgical resection. The positive rates of JADE1 and HBO1 were detected by immunohistochemistry in the resected cancer tissues and paracancerous tissues, and were compared in the cancer tissues of patients with different clinicophathological features. The correlation between JADE1 and HBO1 was evaluated by Spearman correlation analysis. The 3-year overall survival rates were compared between patients with positive and negative JADE1 and HBO1 by Kaplan-Meier survival analysis. Multivariate Cox regression analysis was done to assess the risk factors of poor prognosis of gastric cancer patients. Results JADE1 positive staining was mainly located in the cytoplasm and cell membrane, and HBO1 positive staining was mainly located in the nucleus. The positive rates of JADE1 and HBO1 were significantly higher in the cancer tissues(67.4%, 72.8%) than those in the paracancerous tissues(13.0%, 10.9%)(χ^(2)=56.511, P<0.001;χ^(2)=72.559, P<0.001). There was a positive correlation between JADE1 and HBO1 in the gastric cancer tissues(r=0.491, P<0.001). The positive rates of JADE1 and HBO1 were higher in patients with clinical stage Ⅲ(88.9%, 92.6%) than those in patients with clinical stage Ⅰ-Ⅱ(58.5%, 64.6%)(χ^(2)=8.037, P=0.005;χ^(2)=7.545, P=0.006), higher in patients with lymph node metastasis(87.9%, 90.9%) than those in patients without lymph node metastasis(55.9%, 62.7%)(χ^(2)=9.829, P=0.002;χ^(2)=8.503, P=0.004). The 3-year overall survival rate was lower in patients with positive JADE1(60.7%) and positive HBO1(60.6%) than that in patients with negative JADE1(83.3%) and negative HBO1(88.0%) (χ^(2)=17.562,P<0.001;χ^(2)=18.821,P<0.001).The positive expressions of JADE1(HR=2.141,95%CI:1.293-3.663,P=0.004)and HBO1(HR=3.5
