摘  要:针对丁酸梭菌(Clostridium butyricum)培养环境改善和厌氧发酵的需求,本研究通过采用抽气负压发酵法进行厌氧环境的制造和有害代谢物的排除,以提高发酵效率和芽孢转化率。研究结果显示:相比于传统通氮气正压发酵法,抽气负压发酵法能够创造严格厌氧环境,减少培养环境中甲酸、乙醛、乙醇、丁酸的含量,显著降低有害代谢物及氧气对丁酸梭菌的生长抑制作用。丁酸梭菌菌体浓度达到3.56×10^(9)cfu/mL,芽孢浓度达到3.52×10^(9)cfu/mL,分别较传统通氮气正压法提高了5倍和10倍。本研究为实现丁酸梭菌的高密度发酵提供了有价值的科学指导。In order to achieve the demand for improvement of the culture environment of Clostridium butyricum and the need for anaerobic fermentation,the anaerobic environment is manufactured and harmful metabolites are eliminated by the suctioned negative pressure fermentation to improve the fermentation efficiency and spore conversion rate.The results showed that compared with the traditional positive pressure fermentation with nitrogen,the suctioned negative pressure fermentation can create a strict anaerobic environment,reduce the content of formic acid,acetaldehyde,ethanol,and butyric acid in the culture environment,and significantly reduce harmful metabolites and oxygen inhibits the growth of Clostridium butyricum.The cell concentration reached 3.56×10^(9) cfu/mL,and the spore concentration reached 3.52×10^(9) cfu mL,which were 5 times and 10 times higher than the positive pressure fermentation with nitrogen.This study provides valuable scientific guidance for the realization of high-density fermentation of Clostridium butyricum.
