摘　　要：目的探究Nrf2激活剂RTA-403抗阿尔茨海默病(AD)的药效。方法将实验动物随机分为六组,每组各15只,野生型组(WT组)、Nrf2基因敲除组、颅内注射β淀粉样蛋白组(Aβ组)均给予普通饲料喂养;WT+RTA-403组、Nrf2基因敲除+RTA-403组、Aβ+RTA-403组均给予含有RTA-403[50 mg/(kg·d)]体积分数的饲料喂养,观察各组小鼠行为学和生理生化指标的差异。结果水迷宫潜伏期,Aβ+RTA-403组较Aβ组缩短(P<0.01)。Y迷宫第6周和第8周错误次数,Aβ+RTA-403组低于Aβ组(P<0.05)。小鼠的体重、尿量和血糖,Aβ+RTA-403组高于Aβ组(P<0.05)。水迷宫潜伏期、Y迷宫第6周和第8周错误次数及小鼠的体重、尿量和血糖,WT+RTA-403组较WT组、Nrf2基因敲除+RTA-403组较Nrf2基因敲除组,均无明显变化(P>0.05)。结论Nrf2激活剂RTA-403能显著改善Aβ组小鼠的行为学习能力,Nrf2通路可作为AD治疗的研究方向。Objective To investigate the efficacy of Nrf2 activator RTA-403 against Alzheimer’s disease(AD).Methods Experimental animals were randomly divided into six groups with 15 animals in each group.The wildtype group(WT group),Nrf2 knockout group,and intracranial injection of amyloid-βgroup(Aβgroup)were fed with ordinary feedstuff,while the WT+RTA-403 group,Nrf2 knockout+RTA-403 group,and Aβ+RTA-403 group were all fed with feedstuff containing RTA-403(50 mg/[kg·d])in volume fraction.The differences in ethological indicators and physiological and biochemical indicators were observed in each group.Results Compared with the Aβgroup,the water maze latency shortened in the Aβ+RTA-403 group after intervention(P<0.01).The number of errors in week 6 and week 8 in the Y-maze in the Aβ+RTA-403 group was lower than that in the Aβgroup(P<0.05).The body weight,urine volume and blood glucose of mice in the Aβ+RTA-403 group were higher than those in the Aβgroup(P<0.05).There were no significant changes in the water maze latency,the number of errors at week 6 and week 8 in the Y maze and the body weight,urine volume and blood glucose of mice in the WT+RTA-403 group compared with the WT group,and in the Nrf2 knockout+RTA-403 group compared with the Nrf2 knockout group(P>0.05).Conclusion The Nrf2 activator RTA-403 can significantly improve the behavioral learning ability of mice in the Aβgroup,and the Nrf2 pathway can be a research direction for AD treatment.
