摘　　要：自然杀伤(NK)细胞在对病毒的先天防御以及控制肿瘤生长和转移中发挥着重要作用,NK细胞的免疫效应是由其表面的激活性和抑制性受体所调控的。NKG2A是一种在NK细胞和细胞毒性T淋巴细胞(CD8+T)表面均表达的抑制性受体。在多种肿瘤研究中均发现肿瘤浸润淋巴细胞表面NKG2A表达上调及肿瘤细胞表面其配体人类白细胞抗原E(HLA-E)表达上调。NKG2A与其配体HLA-E的表达上调与肿瘤的免疫逃逸、不良预后有关,且阻断NKG2A可有效缓解NK、CD8^(+)T细胞的免疫学效应,为阻断NKGA/HLA-E轴的单克隆抗体作为新的抗肿瘤免疫治疗的药物研究奠定基础。Natural killer(NK)cells play an important role in innate defense against viruses and in controlling tumor growth and metastasis,and the immune response of NK cells are regulated by activating and inhibitory receptors on their surface.NKG2A is an inhibitory receptor expressed on the surface of both NK cells and cytotoxic T lymphocytes(CD8+T).A variety of tumor studies show that the expression of NKG2A on the surface of tumor-infiltrating lymphocytes is up-regulated,and the expression of its ligand,human leukocyte antigen E(HLA-E),is also up-regulated on the surface of tumor cells.The upregulation of the expressions of NKG2A and its ligand HLA-E is found to be associated with immune escape and poor prognosis of tumors,and blockade of NKG2A effectively alleviates the immunological effects of NK and CD8^(+)T cells,which provide a basis for the study of monoclonal antibodies that block the NKGA/HLA-E axis,to be new agents for anti-tumor immunotherapy.
