摘 要:从黄芫花甲醇提取液中制备获得一富含8个黄酮的黄芫花提取物。8个黄酮经NMR分析结构鉴定为:isoorientin,orientin,vitexin,isovitexin,quercetin 3-O-β-glucopyranoside,cynaroside,astragalin和luteolin-4′-O-glucoside。进一步构建了同时测定该提取物中8个黄酮含量的HPLC方法,方法学考察表明8个黄酮化合物有很好的分离效果,线性关系、稳定性、重复性较好。3批黄芫花黄酮提取物中含量测定结果显示提取物中8个黄酮化合物总含量为(55.19±0.32)%。FRAP和DPPH抗氧化测定结果显示8个黄酮合物及总黄酮提取物FRAP值为0.19μmol/mL~0.84μmol/mL,DPPH·清除率的IC_(50)值4.03μg/mL~149.56μg/mL。文章不仅揭示了黄芫花中一类重要的药效物质,更重要的是所获得的黄芫花黄酮提取物制备容易、含量稳定、抗氧化能力强,具有广泛的应用前景。A flavonoid extracts including eight flavonoid compounds was prepared by LH-20 column chromatography from the bud of Wikstroemia chamaedaphne. The structure of the 8 flavonoid compounds was isolated by chromatographic and spectral techniques: isoorientin, orientin, vitexin, isovitexin, quercetin 3-O-β-glucopyranoside, cynaroside, astragalin, and luteolin-4’-O-glucoside. Moreover, a HPLC method for simultaneously determining the content of 8 flavonoids in the extract was further constructed,and methodological investigation showed that the 8 flavonoid compounds in this method had a good separation effect, and the linear relationship, stability, and repeatability were good. The total content of 8 flavonoid compounds was(55. 19±0. 32)% in the flavonoid extracts. The FRAP values and the IC_(50) of DPPH free radical-scavenging for the 8 flavonoid compounds and the flavonoid extracts were 0. 19 μmol/mL-0. 84 μmol/mL and 4. 03 μg/mL-149. 56 μg/mL, respectively. On the one hand, this study reveals an important class of medicinal substances in the bud of W. chamaedaphne. On the other hand, the flavonoid extracts from the bud of W. chamaedaphne is easy to prepare, stable in content, and has strong antioxidant activity, which shows a wide range of application prospects.
