摘  要:目的探讨没食子酸(gallic acid, GA)对小鼠骨髓细胞的辐射防护作用。方法将健康雄性ICR小鼠随机分为生理盐水对照组、GA对照组、4个X线照射组和4个GA保护组,每组10只。X线照射组和生理盐水对照组给予0.01 mL/g生理盐水灌胃,GA对照组和GA保护组给予200 mg/kg的GA(20 mg/mL)灌胃14 d,第15 d给予4个X线照射组和4个GA保护组小鼠全身一次性X线照射,吸收剂量分别为1.0、2.0、3.0和4.0 Gy,生理盐水对照组和GA对照组不给予照射。照射后检测全血过氧化氢酶(catalase, CAT)、超氧化物歧化酶(superoxide dismutase, SOD)、丙二醛(malondialdehyde, MDA)和骨髓嗜多染红细胞微核(micronucleus frequency of polychromatic erythrocyte, MN-PCE)率,并用流式细胞仪检测骨髓细胞周期、早期凋亡率和晚期凋亡率。结果 1.0、2.0、3.0和4.0 Gy GA保护组小鼠血清中CAT活性分别为2.13、1.74、1.49和1.15 U/mL,与相应X线照射组比较均明显增加(P<0.01);SOD活性分别为184.69、156.92、139.17和107.15 U/mL,与相应X线照射组比较均明显增加(P<0.01);MDA含量分别为3.92、4.20、6.32和9.31 nmol/mL,与相应X线照射组比较均明显降低(P<0.05,P<0.01);1.0、2.0、3.0和4.0 Gy GA保护组小鼠骨髓MN-PCE率分别为4.35‰、8.00‰、12.90‰和3.80‰,与相应X线照射组比较均明显降低(P<0.01);1.0、2.0、3.0和4.0 Gy GA保护组小鼠骨髓细胞G;/G;期细胞所占比例分别为81.00%、86.28%、92.04%和93.15%,与相应X线照射组比较均明显减少(P<0.01);G;/M期细胞所占比例分别为4.51%、3.05%、2.35%和1.81%,与相应X线照射组比较均明显增加(P<0.05,P<0.01);S期细胞所占比例分别为15.32%、11.36%、5.96%和4.92%,与相应X线照射组比较均明显增加(P<0.01)。1.0、2.0、3.0和4.0 Gy GA保护组小鼠骨髓细胞早期凋亡率分别为3.32%、8.96%、12.11%和2.26%,与相应X线照射组比较均明显减少(P<0.01);骨髓细胞晚期凋亡率分别为7.21%、11.73%、17.11%和19.36%,与相应X线照�OBJECTIVE To investigate the radioprotective effect of gallic acid(GA) on mouse bone marrow cells. METHODS Healthy male ICR mice were randomly divided into saline control group, GA control group, X-ray irradiation group and GA protection group, with 10 mice in each group. X-ray irradiation group and normal saline control group were given 0.01 mL/g normal saline gavage, GA control group and GA protection group were given 200 mg/kg GA(20 mg/mL) gavage once a day for 14 consecutive days. On the 15 th day, 4 X-ray irradiation groups and 4 GA protection groups were given one-time X-ray irradiation to the whole body of the mice, and the absorbed doses were 1.0, 2.0, 3.0 and 4.0 Gy, respectively. The saline control group and the GA control group were not irradiated. After irradiation, detected the whole blood catalase(CAT), superoxide dismutase(SOD), malondialdehyde(MDA) and micronucleus frequency of polychromatic erythrocyte in bone marrow(MN-PCE), and use flow cytometry to detect bone marrow cell cycle, early apoptosis rate and late apoptosis rate. RESULTS The CAT activities in the serum of mice in the 1.0, 2.0, 3.0 and 4.0 Gy GA protection groups were 2.13, 1.74, 1.49 and 1.15 U/mL, respectively, which were significantly increased compared with the corresponding X-ray irradiation group(P<0.01). SOD activities were 184.69, 156.92, 139.17 and 107.15 U/mL, which were significantly increased compared with the corresponding X-ray irradiation group(P<0.01). The contents of MDA were 3.92, 4.20, 6.32 and 9.31 nmol/mL, which were significantly lower than those of the corresponding X-ray irradiation group(P<0.05, P<0.01). The bone marrow MN-PCE rate of the mice in the 1.0, 2.0, 3.0 and 4.0 Gy GA protection groups were 4.35‰, 8.00‰, 12.90‰ and 3.80‰, respectively, which were significantly lower than those in the corresponding X-ray irradiation group(P<0.01). The proportions of G;/G;phase cells of bone marrow cells in the 1.0, 2.0, 3.0 and 4.0 Gy GA protection group were 81.00%, 86.28%, 92.04% and 93.15%, respectively, w
