摘  要:目的:探讨CCAAT/增强子结合蛋白-α(CEBPA)在肝细胞肝癌(HCC)中的表达及其临床意义。方法:收集TCGA、GTEx和GEO等外文数据库中CEBPA在肝细胞肝癌中的表达值,对3727例癌样本和3204例非癌样本进行综合分析,用实时定量逆转录聚合酶链反应技术验证在肝细胞癌细胞系(Huh7)和正常肝细胞细胞系(L02)中CEBPA的表达,免疫组化技术验证90例肝细胞癌组织和90例癌旁组织CEBPA的表达情况。结果:公共数据集分析结果显示,CEBPA在癌组织中的表达明显高非癌组织(SMD=0.51),实验验证CEBPA在Huh7肝细胞癌细胞系中的表达明显高于正常肝细胞细胞系(P<0.01)。CEBPA蛋白的差异表达在HCC患者病理分级及TNM分期中的T分期有显著差异(P<0.05)。将HCC中CEBPA相关基因、差异基因和靶基因的交叉基因进行的KEGG分析,发现这些基因富集在细胞周期等通路上。结论:CEBPA在肝细胞肝癌组织中表达较高,可能参与肝细胞肝癌的发生发展。Objective:To investigate the expression of CCAAT/enhancer binding protein-α(CEBPA)and its clinical significance in hepatocellular carcinoma(HCC).Methods:The expression values of CEBPA in HCC were collected from TCGA,GTEx and GEO databases.A total of 3,727 cancer samples and 3,204 non-cancer samples were collected were comprehensively analyzed.The expression of CEBPA in hepatocellular carcinoma cell line(Huh7)and normal hepatocellular cell line(L02)was verified by RT-qPCR.Immunohistochemical technique was used to verify the expression of CEBPA in 90 hepatocellular carcinoma tissues and 90 para-tumor groups.Results:Public data set analysis showed that the expression of CEBPA in cancer tissues was significantly higher than that in non-cancer tissues(SMD=0.51),and it was verified that the expression of CEBPA in Huh7 hepatocellular carcinoma cell line was significantly higher than that in normal liver cell line(P<0.01).The differential expression of CEBPA protein was statistically different in pathological grade and the T stage of TNM stage for HCC patients(P<0.05).KEGG analysis of CEBPA CEBPA-related genes,differential genes and cross genes of target genes in HCC showed that they were enriched in cell cycle and other pathways.Conclusion:CEBPA is highly expressed in hepatocellular carcinoma tissuesand may be involved in the occurrence and development of hepatocellular carcinoma.
