摘  要:目的:探讨黑加仑提取物对肺癌细胞A549的细胞周期、凋亡和迁移的影响及可能机制。方法:体外培养A549细胞,分为对照组、不同浓度黑加仑提取物组(5 mg/mL、10 mg/mL、15 mg/mL)、pcDNA组、pcDNA-PSMA3-AS1组、15 mg/mL黑加仑提取物+pcDNA组、15 mg/mL黑加仑提取物+pcDNA-PSMA3-AS1组。流式细胞术检测细胞周期和凋亡;划痕实验检测细胞迁移;western blotting法检测细胞中Bcl-2和Bax蛋白表达;RT-qPCR法检测PSMA3-AS1表达。结果:与对照组比较,10 mg/mL、15 mg/mL黑加仑提取物组A549细胞周期G0~G1期延长,S期缩短,细胞凋亡率和Bax蛋白表达升高,而划痕愈合率和Bcl-2蛋白、PSMA3-AS1表达降低(均P<0.05)。与pcDNA组比较,pcDNA-PSMA3-AS1组A549细胞周期G0~G1期缩短,S期延长,凋亡率和Bax蛋白表达降低,而划痕愈合率和Bcl-2蛋白表达升高(均P<0.05)。与pcDNA-PSMA3-AS1组比较,15 mg/mL黑加仑提取物+pcDNA-PSMA3-AS1组A549细胞凋亡率和细胞中Bax蛋白表达升高,划痕愈合率和Bcl-2蛋白、PSMA3-AS1表达降低(均P<0.05)。与15 mg/mL黑加仑提取物+pcDNA组比较,15 mg/mL黑加仑提取物+pcDNA-PSMA3-AS1组A549细胞周期G0~G1期缩短,S期延长,细胞凋亡率和细胞中Bax蛋白表达降低,划痕愈合率和Bcl-2蛋白、PSMA3-AS1表达升高(均P<0.05)。结论:黑加仑提取物可能通过下调PSMA3-AS1表达阻滞肺癌细胞周期进程,促进细胞凋亡,并抑制细胞迁移。Objective:To investigate the effects and possible mechanisms of blackcurrant extract on the cell cycle,apoptosis and migration of lung cancer cells A549.Methods:A549 cells were cultured in vitro and divided into control group,different concentration of blackcurrant extract(5 mg/mL,10 mg/mL,and 15 mg/mL)groups,pcDNA group,pcDNA-PSMA3-AS1 group,15 mg/mL blackcurrant extract+pcDNA group,and 15 mg/mL blackcurrant extract+pcDNA-PSMA3-AS1 group.Flow cytometry was used to detect cell cycle and apoptosis.Scratch test was used to detect cell migration.Western blotting was used to detect the protein expressions of Bcl-2 and Bax.RT-qPCR was used to detect the expression of PSMA3-AS1.Results:Compared with the control group,the G0-G1 phase of A549 cell cycle in 10 mg/mL and 15 mg/mL blackcurrant extract groups were prolonged,the S phase were shortened,and the apoptosis rate and the protein expression of Bax were increased,while scratch healing rate and the expressions of Bcl-2 protein and PSMA3-AS1 were decreased(P<0.05).Compared with the pcDNA group,the G0-G1 phase of A549 cells in the pcDNA-PSMA3-AS1 group was shortened,the S phase was prolonged,and the apoptosis rate and the protein expression of Bax were decreased,whereas scratch healing rate and the expressions of Bcl-2 protein and PSMA3-AS1 were increased(P<0.05).Compared with the pcDNA-PSMA3-AS1 group,the apoptosis rate and the protein expression of Bax were increased in the 15 mg/mL blackcurrant extract+pcDNA-PSMA3-AS1 group,but scratch healing rate and the expressions of Bcl-2 protein and PSMA3-AS1 were decreased(P<0.05).Compared with the 15 mg/mL blackcurrant extract+pcDNA group,the G0-G1 phase of A549 cells in the 15 mg/mL blackcurrant extract+pcDNA-PSMA3-AS1 group was shortened,the S phase was prolonged,and the apoptosis rate and the protein expression of Bax decreased,but scratch healing rate and the expressions of Bcl-2 protein and PSMA3-AS1 increased(P<0.05).Conclusion:Blackcurrant extract may block the cell cycle progression of lung cancer cells,promote cell a
