摘 要:目的:研究维生素E对慢性癫痫大鼠海马组织脂氧合酶15(15-LOX)和氧化应激的影响.方法:选用雄性SD大鼠36只,随机分为3组,分别为对照组、癫痫组和维生素E处理组.采用戊四氮(PTZ)诱导的慢性癫痫大鼠模型并用维生素E干预,通过行为学观察大鼠癫痫发作程度,运用全细胞膜片钳技术分析大鼠海马神经元动作电位的变化情况,采用分光光度计法检测海马组织中丙二醛(MDA)和谷胱甘肽(GSH)含量,应用免疫荧光染色和Western Blot检测15-LOX表达情况.结果:与对照组相比,癫痫组大鼠癫痫发作明显,海马区MDA含量和15-LOX表达升高,GSH含量降低,神经元兴奋性升高(P<0.01);与癫痫组相比,维生素E处理组大鼠癫痫发作强度降低,海马区MDA和15-LOX表达明显降低,GSH含量升高,神经元兴奋性降低(P<0.01).结论:维生素E可通过抑制15-LOX降低氧化应激和神经元的兴奋性进而影响癫痫发作.Objective:To study the effects of vitamin E on 15-lipoxygenase(15-LOX)and oxidative stress in the hippocampus of rats with chronic epilepsy.Methods:Thirty-six male Sprague-Dawley rats were randomly divided into three groups:control group,epilepsy group and vitamin E treatment group.The chronic epileptic rat model induced by pentylenetetrazol(PTZ),and intervened with vitamin E.The seizure degree was observed by behavior,the changes of the action potential in hippocampal neurons were analyzed by whole-cell patch-clamp technique,the contents of malond-ialdehyde(MDA)and glutathione(GSH)in hippocampal tissue were detected by spectrophotometer,and the expression of 15-LOX was detected by immunofluorescence staining and Western Blot.Results:It’s more obvious of epileptic seizure for rats in epilepsy group compared to the control group,the content of MDA and expression of 15-LOX increased,the level of the GSH decreased and the excitability of hippocampal neurons increased significantly(P<0.01);However,when rats were injected with vitamin E compared with epilepsy group,the seizure intensity decreased,the content of MDA and expression of 15-LOX decreased obviously,the level of the GSH increased and the excitability of hippocampal neurons decreased significantly(P<0.01).Conclusion:Vitamin E can reduce oxidative stress and neuronal excitability by inhibiting 15-LOX.
