摘 要:目的 建立新型斑马鱼的人口腔鳞状细胞癌(OSCC)药物筛选模型,并在体内实时动态观察山竹醇(Garcinol)对OSCC细胞的影响。方法 培养转基因斑马鱼Tg(fli1a:EGFP),不同浓度的Garcinol处理胚胎5天,筛选最适浓度。培养人OSCC细胞系SCC-15,不同浓度的细胞膜红色荧光探针(DiI)标记细胞,筛选最适浓度。通过显微注射建立斑马鱼OSCC模型。通过Garcinol药物实验,实时动态观察0.5μmol/L、1μmol/L Garcinol处理24 h和48 h后对体内OSCC细胞增殖的影响。结果 利用斑马鱼初步建立一种新型的OSCC药物筛选模型,并发现天然产物Garcinol能明显抑制斑马鱼体内OSCC细胞的增殖,其抑制能力呈一定的浓度依赖性。结论 Garcinol对OSCC细胞增殖有明显的抑制作用,有作为治疗口腔癌药物的应用前景。此外,斑马鱼OSCC药物筛选模型可以应用于抗口腔癌药物筛选的初步研究。Objective To establish a novel zebrafish human oral squamous cell carcinoma(OSCC) drug screening model and observe the effect of Garcinol on OSCC cells. Methods The zebrafish transgenic line Tg(fli1 a: EGFP) was cultured, and embryos were treated with Garcinol at different concentrations for 5 days to screen the optimal concentration.Human OSCC cell line SCC-15 was cultured, and cells were labeled with red fluorescent probe(DiI) at different concentrations to screen the optimal concentration. The zebrafish OSCC model was established by microinjection experiment.The effect of 0.5 μmol/L and 1 μmol/L Garcinol on the proliferation of OSCC cells was observed in vivo after 24 hours and 48 hours. Results OSCC drug screening model was established in zebrafish body. Garcinol could inhibit the proliferation of OSCC cells in zebrafish body in a concentration dependent manner. Conclusions Garcinol has an inhibitory effect on the proliferation of OSCC cells.
