摘 要:目的研究miR-154靶向HMGA2基因在人子宫内膜癌中的表达及其对子宫内膜癌细胞恶性生物学行为的影响。方方法检测各组样本中HMGA2 mRNA、miR-154的表达水平和HMGA2蛋白表达水平。培养人子宫内膜癌细胞株HEC-1-B、HHUA、hEEC和正常子宫内膜细胞株hESC,构建miR-154模拟物,检测各组细胞中HMGA2蛋白、miR-154及HMGA2 mRNA的表达情况,荧光素酶报告实验鉴定HMGA2与miR-154的靶向关系,检测各组细胞的增殖、迁移和侵袭情况。结果HMGA2 mRNA和蛋白在子宫内膜癌中高表达,miR-154在子宫内膜癌中低表达。miR-154的表达量与子宫内膜疾病的严重程度呈负相关关系,HMGA2的表达量与子宫内膜疾病的严重程度呈正相关关系。HMGA2基因的表达显著提高了子宫内膜癌细胞的增殖、迁移与侵袭能力(P<0.05)。结论miR-154可通过负调控HMGA2抑制子宫内膜癌细胞的恶性生物学行为。Objective To study the expression of miR-154 targeting HMGA2 gene in human endometrial cancer and its effect on the malignant biological behavior of endometrial cancer cells.Methods The expression levels of HMGA2 mRNA,miR-154 and the protein level of HMGA2 in each group of patient samples were detected.Human endometrial cancer cell lines HEC-1-B,HHUA,hEEC and normal endometrial cell lines hESC were cultured,and miR-154 mimics were constructed.The expression of HMGA2 protein,miR-154 and HMGA2 mRNA in each group of cells were detected.The luciferase reporter assay was used to identify the targeting relationship between HMGA2 and miR-154.The proliferation,migration and invasion of cells in each group were detected.Results The expression levels of HMGA2 mRNA and protein were highly expressed in endometrial carcinoma,miR-154 was low expressed in endometrial carcinoma.The expression of miR-154 was negatively correlated with the severity of endometrial disease.There was a positive correlation between the expression of HMGA2 and the severity of endometrial disease.The expression of HMGA2 gene significantly improved the proliferation,migration and invasion ability of endometrial cancer cells(P<0.05).Conclusion miR-154 can inhibit the malignant biological behavior of endometrial cancer cells by negatively regulating HMGA2.
