摘  要:目的进一步研究胃癌发病机制。方法富集分析3个GEO数据集筛选出的差异基因,利用STRING数据库绘制蛋白质相互作用图,使用Cytoscape软件筛选关键基因。通过TCGA数据库验证关键基因的差异表达。利用Western blotting和实时荧光定量PCR(qPCR)检测胃癌组织中细胞周期相关基因的蛋白和mRNA的表达水平。结果共筛选出203个差异表达基因,其中包括131个上调表达和72个下调表达基因。与癌旁组织相比,周期蛋白依赖性激酶1(CDK1)、细胞周期蛋白B1(CCNB1)和有丝分裂停滞缺陷蛋白2(MAD2L1)在胃癌组织中明显上调表达(P<0.05)。在胃癌组织中,CCNB1和CDK1之间存在显著正相关关系(R=0.98,P<0.001)。Western blotting和qPCR结果显示,胃癌组织中CDK1、CCNB1和MAD2L1的蛋白和mRNA水平均上调表达(P<0.05)。结论CDK1、CCNB1和MAD2L1在胃癌组织中上调表达,有可能成为治疗胃癌的新的潜在生物标志物。Objective To further study the pathogenesis of gastric cancer(GC).Methods Enrichment analysis was performed on the differential genes selected from the three GEO datasets,protein-protein interaction network was drawn using the STRING database,the key genes were screened using Cytoscape software.The differential expression of key genes was verified by TCGA database.Western blotting and real-time quantitative PCR(qPCR)were used to detect the protein and mRNA expression levels of cell cycle-related genes in GC tissues.Results 203 differentially expressed genes were screened,including 131 up-regulated genes and72 down-regulated genes.Compared with adjacent tissues,cyclin-dependent kinase 1(CDK1),cyclin B1(CCNB1)and mitotic arrest defect protein 2 like 1(MAD2 L1)were significantly up-regulated in GC tissues(P<0.05).There was a significant positive correlation between CCNB1 and CDK1 in GC tissues(R=0.98,P<0.001).Western blotting and qPCR results showed that the protein and mRNA levels of CDK1,CCNB1 and MAD2 L1 were up-regulated in GC tissues(P<0.05).Conclusion CDK1,CCNB1 and MAD2 L1,when up-regulated in GC tissues,may become new potential biomarkers for the treatment of GC.
