摘  要:结节性硬化症(TSC)是一种常染色体显性遗传性疾病。TSC是神经皮肤综合征中相对常见的一种,其临床特征可出现脑、皮肤、周围神经、肾等多器官受累,面部血管纤维瘤、癫痫发作和智能减退。癫痫是最常见的、严重的且对治疗耐受的疾病之一。既往抗癫痫发作治疗主要控制性发作症状,减少癫痫发作的频率和(或)减轻发作程度但并无持久的抗癫痫作用(即不改善疾病本身),也无法预防和延缓癫痫的发展(即不阻止癫痫的产生),更无法预防和减轻癫痫共患病的产生。疾病修饰疗法(DMT)是通过医学干预达到改变疾病临床进展轨迹的治疗方法。癫痫DMT主要目的在于阻止或逆转癫痫的进展,或者阻止易感人群发病。文中对癫痫DMT在结节性硬化症哺乳类动物雷帕霉素靶蛋白方面的研究进展进行综述。Previously, anti-epilepsy drugs were mainly used to control the symptoms of seizures,reducing the frequency and/or severity degree of seizures, without preventing epileptic and delaying the progresses of epilepsy, and without preventing or reducing the occurrence of epilepsy comorbidities.Disease modification therapy(DMT) is a treatment method that changes the course of disease by medical intervention to prevent or reverse the progression of epilepsy, or prevent the onset of epilepsy in susceptible people. The research progresses of DMT for epilepsy in the field of mammalian target of rapamycin of tuberous sclerosis complex were reviewed.
