摘  要:目的探讨氧化苦参碱(Oxymatrine)联合紫杉醇对人胃癌细胞HGC-27和裸鼠移植瘤生长的影响。方法体外培养人胃癌细胞HGC-27,将对数期的HGC-27细胞随机分为对照组(不处理)、氧化苦参碱组(氧化苦参碱2 mg/ml)、紫杉醇组(紫杉醇10μmol/ml)和氧化苦参碱联合紫杉醇组(氧化苦参碱2 mg/ml+紫杉醇10μmol/ml),4组经相应处理后采用噻唑蓝比色法(MTT)检测细胞增殖情况,流式细胞术检测细胞凋亡率,蛋白免疫印迹(Western blot)分析增殖和凋亡相关蛋白增殖细胞核抗原(PCNA)和剪切的半胱氨酸蛋白酶3(Cleaved Caspase-3)的表达。构建人胃癌HGC-27细胞裸鼠移植瘤模型28只,随机分为4组:对照组(Mock组:0.9%氯化钠溶液)、氧化苦参碱组(Oxy组:氧化苦参碱75 mg/kg)、紫杉醇组(PTX组:紫杉醇10 mg/kg)和氧化苦参碱联合紫杉醇组(Oxy+PTX组:氧化苦参碱75 mg/kg+紫杉醇10 mg/kg),每组7只。4组连续用药28 d,颈椎脱臼处死裸鼠,测量肿瘤体积和质量,计算抑瘤率。结果人胃癌细胞HGC-27体外实验结果显示,与对照组相比,氧化苦参碱组和氧化苦参碱联合紫杉醇组HGC-27细胞的存活率和PCNA的表达明显降低(P<0.05),凋亡率和Cleaved Caspase-3的表达明显升高(P<0.05);且氧化苦参碱组和紫杉醇组与氧化苦参碱联合紫杉醇组相比,差异均有统计学意义(P<0.05)。裸鼠移植瘤实验结果显示,Oxy+PTX组抑瘤率明显高于Mock组、Oxy组和PTX组(P<0.05)。结论氧化苦参碱和紫杉醇联合使用能够协同抑制人胃癌细胞HGC-27和裸鼠移植瘤的生长。Objective To investigate the effects of oxymatrine combined with paclitaxel on the growth of human gastric cancer cell HGC-27 and transplanted tumors in nude mice.Methods Human gastric cancer cells-HGC-27 were cultured in vitro,and HGC-27 cells in Log phase were randomly divided into control group(no treatment),oxymatrine group(oxymatrine 2mg/ml),paclitaxel group(paclitaxel 10μmol/ml)and oxymatrine combined with paclitaxel group(oxymatrine 2mg/ml+paclitaxel 10μmol/ml).After corresponding treatment,the four groups were treated with thiazole blue colorimetry(MTT)to detect cell proliferation,and flow cytometry was used to detect the rate of cell apoptosis.Western Blot was used to analyze the expression of proliferation and apoptosis-related proteins,proliferating cell nuclear antigen(PCNA)and cleaved Caspase 3.Twenty-eight nude mice transplanted tumor models of human gastric cancer HGC-27 cell were constructed and randomly divided into 4 groups:control group(Mock:0.9%sodium chloride solution),oxymatrine group(Oxy:oxymatrine 75mg/kg),paclitaxel group(PTX:paclitaxel 10mg/kg)and oxymatrine combined with paclitaxel group(Oxy+PTX:oxymatrine 75mg/kg+paclitaxel 10mg/kg),with 7 mice in each group.The four groups were treated for 28 consecutive days,and the nude mice were sacrificed by cervical dislocation.The tumor volume and mass were measured,and the tumor inhibition rate was calculated.Results The results of in vitro experiments showed that compared with those in Mock group,the survival rate and PCNA expression levels of HGC-27 cells in Oxy group and Oxy+PTX group were significantly decreased(P<0.05),however,the apoptosis rate and the expression levels of Cleaved Caspase 3 were significantly increased(P<0.05),there were significant differences between Oxy group,PTX group and Oxy+PTX group(P<0.05).The experimental results of transplanted tumors in nude mice showed that the tumor inhibition rate in Oxy+PTX group was significantly higher than that in Mock group,Oxy group and PTX group(P<0.05).Conclusion The combined appli
