摘  要:为研究SDC-1的细胞定位对人肝癌细胞侵袭能力的影响,利用HPSE小干扰RNA和HPSE抑制剂低分子量肝素处理高转移人肝癌细胞MHCC97-H,通过实时定量PCR方法检测HPSE的mRNA表达,应用免疫细胞化学方法分析SDC-1的细胞定位,通过基质胶侵袭实验观察细胞侵袭性的改变。结果显示:HPSE小干扰RNA能够显著干预HPSE的mRNA表达,低分子量肝素对HPSE的mRNA表达没有影响;HPSE表达及活性下调后细胞表面的SDC-1明显增加,并能有效抑制细胞的体外侵袭能力。结果表明,肝癌细胞表面定位的SDC-1是阻断肝癌细胞转移的重要因素。To study the effect of cell localization of SDC-1 on invading human hepatocellular carcinoma cells,MHCC97-H cells were treated with HPSE small interfering RNA and low molecular weight heparin(HPSE inhibitor).The mRNA expression of HPSE was detected by real-time quantitative PCR.The cell localization of SDC-1 was analyzed by immunocytochemistry.The changes of cell invasion were observed by matrix gel invasion assay.The results showed that HPSE small interfering RNA could significantly interfere with HPSE mRNA expression,and low molecular weight heparin had no effect on HPSE mRNA expression.After the down-regulatiing HPSE expression and activity,SDC-1 on the cell surface significantly increased,and the in vitro invasion ability of cells effectively inhibited.The results showed that SDC-1 localized on the surface of HCC cells was an important factor in blocking the metastasis of HCC cells.
