摘  要:目的:探究ERK抑制剂U0126对氧糖剥夺再灌注致神经母细胞瘤(SH-SY5Y)细胞损伤的影响。方法:将生长状态良好的SH-SY5Y细胞随机分为正常对照组(Control组)、模型组(OGD/R组)、溶剂组(DMSO组)、U0126 10μmol/L组(U0126-L组)、U0126 20μmol/L组(U0126-M组)和U0126 30μmol/L组(U0126-H组)。U0126作用24 h后,除Control组更换正常完全培养基外,其余组采用DMEM无糖培养基和氮气缺氧法制备SHSY5Y细胞缺糖缺氧2 h后复糖复氧,继续放入37℃、5%CO2培养箱中培养24 h。采用CCK-8法测定细胞存活率;酶标法检测细胞培养液乳酸脱氢酶(LDH)活性和细胞超氧化物歧化酶(SOD)活性;流式细胞术测定细胞凋亡率。结果:与OGD/R组比较,Control组、U0126三个剂量组的LDH活性和细胞凋亡率均明显降低,而细胞中SOD活性显著升高(P<0.05);U0126作用显示出剂量效应关系,U0126-H组与U0126-L组有显著性差异(P<0.05)。结论:U0126对SY-SY5Y细胞缺糖缺氧再灌注损伤有一定的保护作用,可提高细胞存活率、增强细胞的抗氧化能力和降低细胞凋亡率。Objective:To investigate the effect of ERK inhibitor U0126 on neuroblastoma(SH-SY5 Y)cell damage induced by oxygen and glucose deprivation reperfusion.Methods:SH-SY5 Y cells were randomly divided into normal Control group(Control group),model group(OGD/R group),solvent group(DMSO group),U0126 10 μmol/L group(U0126-L group),U0126 20 μmol/L group(U0126-M group)and 30 μmol/L group(U0126-H group).After U0126 treatment for 24 h,SH-SY5 Y cells were treated with DMEM glucose-free medium and nitrogen for 2 h,and then reoxygenated and reoxygenated in an incubator with 5% CO2 at 37 ℃ for 24 h,except for Control group,which replaced normal complete medium.Cell viability was determined by CCK-8 method.Lactate dehydrogenase(LDH)activity in cell culture medium and superoxide dismutase(SOD)activity of cells were detected by enzymolysis assay.The apoptosis rate was determined by flow cytometry.Results:Compared with OGD/R group,LDH activity and apoptosis rate in Control group and three U0126 groups were significantly decreased,while SOD activity was significantly increased(P<0.05);The effect of U0126 showed a dose-response relationship,and there was a significant difference between U0126-H and U0126-L groups(P<0.05).Conclusion:U0126 can protect SY-SY5 Y cells from oxygen-glucose deprivation/reperfusion injury,improve cell survival rate,enhance antioxidant capacity and reduce apoptosis rate.
