我国部分地区黄羽肉鸡群鸡传染性贫血病病毒的流行病学调查及致病性分析

摘　　要：为调查鸡传染性贫血病病毒(CIAV)在我国黄羽肉鸡群中的流行情况及毒株致病性情况,于2021年1—8月对山东、江苏、广东、安徽等10个省市417个黄羽肉鸡群CIAV感染情况进行了调查,每个鸡群采集5只病弱鸡肝脏样品,混合后进行PCR检测;同时对一CIAV阳性种鸡群进行了解剖和病理学诊断,并进行了CIAV分离、测序和致病性分析。结果显示:417个鸡群中CIAV阳性群181个,阳性群占比为43.41%;疑似发病种鸡表现冠、面部皮肤苍白,血液吉姆萨染色可见幼稚型红细胞,剖检和病理学诊断均可见骨髓脂肪化;全基因组测序显示分离株具有高致病性毒株的分子特征,与NCBI已发表的其他毒株进行遗传进化分析,结果显示与亚洲CIAV遗传关系相近;1日龄SPF鸡经肌肉注射途径接种分离病毒,接种鸡出现明显的胸部皮炎、骨髓脂肪化等发病症状,死亡率达87.5%,表明分离的CIAV野毒株具有较强致病性。研究结果为当前CIAV研究和疾病的防控提供了参考数据。In order to investigate the epidemiological characteristics and pathogenicity of chicken infectious anemia virus(CIAV)in yellow-feather chickens in China,an epidemiological survey of CIAV,carried out by PCR detection of sample mixtures of five liver from each chicken flock,was conducted in 417 yellow-feather chicken flocks of ten provinces including Shandong,Jiangsu,Guangdong,Anhui and other provinces,from January to August,2021.Anatomic and pathological diagnosis of a CIAV positive breeding flock were also carried out,as well as the isolation,sequencing,and virulence analysis of the CIAV.The results showed that,181 of 417 chicken groups were detected CIAV positive,accounting for 43.41%.The crown and facial skin of suspected diseased breeding chickens showed pale,while immature red blood cells were observed in blood by Giamsa staining,and the marrow fatty conversion was also observed in necropsy and pathological diagnosis.Molecular characteristics of high virulence and close genetic relationship to Asian CIAV could be found by genome sequence analysis and genetic evolution analysis with other CIAV sequences published on NCBI.1-day-old SPF chickens were inoculated with isolated virus by intramuscular injection,and obvious symptoms such as chest dermatitis and bone marrow fatty conversion were found in inoculated chickens,as well as the mortality rate up to 87.5%,indicating that the pathogenicity of isolated CIAV was high.This study provided a reference data for the study,prevention and control of CIAV.
