摘 要:目的:探讨早孕小鼠子宫内膜组织中P53凋亡刺激蛋白抑制因子(IASPP)的表达对胚胎着床的作用。方法:对NIH雌鼠进行阴道涂片以确定其动情周期,采用实时荧光定量PCR、Western blot以及免疫组化等分别在分子和蛋白水平上检测雌鼠子宫内膜上IASPP的表达情况;对实验组小鼠左侧子宫角注射IASPP抑制剂,最后测量IASPP在未孕小鼠及妊娠小鼠第2至7天子宫内膜的表达和胚胎着床数,进行统计分析。结果:IASPP的转录水平随时间推移而缓慢升高,并于雌鼠孕期内的第4天达到峰值(P<0.05),之后IASPP水平缓慢降低。Western blot实验结果也表明IASPP的蛋白水平随时间推移而缓慢增加,在雌鼠动情周期的第5天到达峰值(P<0.05),随后缓慢降低,与转录水平变化结果大致相近。免疫组织化学染色的结果显示,早孕小鼠子宫腔上皮、腺上皮的胞膜和胞浆,血管内皮以及基质细胞的细胞核表达IASPP蛋白均高于未孕组,且各个妊娠阶段表达存在差异;从怀孕开始,IASPP呈现表达逐渐增强的趋势,至孕第7天表达开始下降。从孕第8天开始,实验组左侧子宫胚胎着床数为(1.402±0.517),右侧为(6.420±1.075),对照组左侧子宫胚胎着床数的范围为(6.500±1.090),对照组右侧子宫胚胎着床数的范围为(6.200±0.919),数据显示实验组小鼠的左侧胚胎着床数明显低于右侧胚胎着床数(P<0.05),而对照组小鼠的左右侧子宫胚胎着床数目无明显差别(P>0.05)。结论:妊娠早期的小鼠子宫内膜组织中p53凋亡刺激蛋白抑制因子IASPP持续表达,并正向参与胚胎的着床调控过程。Objective:To investigate the expression effect of inhibitor of apoptosis-stimulating protein P53(IASPP)on embryo implantation in the endometrial tissue of early-pregnant mice.Methods:Vaginal smears were performed on NIH female mice to determine their estrous cycles.The expression of IASPP on endometrium of female mice was detected at the molecular and protein levels by real-time fluorescence quantitative PCR,Western-blot and immunohistochemistry experiments,respectively.The left uterus of the mice in the experimental group was injected with IASPP inhibitor.Eventually,the expression of IASPP in the endometrium and the number of embryo implantation on the 2 nd,3 rd,4 th,5 th,6 th and 7 th days of non-pregnant/pregnant mice were measured and statistically analyzed.Results:The transcription level of IASPP increased slowly over time and reached the peak on the 4 th day of pregnancy in female mice(P<0.05),then decreased slowly.Similarly,the results of Western-blot also showed that the protein level of IASPP increased slowly over time,reaching a peak on the 5 th day of the estrous cycle of female mice(P<0.05)then decreasing slowly.The results of immunohistochemical staining demonstrated that the expression of IASPP in the uterine cavity epithelium,glandular epithelium,vascular endothelium and nucleus of stromal cells of early-pregnant mice was higher than that of non-pregnant group,with differences in the expression of each pregnancy stage.At the beginning of pregnancy,the IASPP expression gradually increased,and began to decline on the seventh day of pregnancy.The number(mean±SD)of embryo implantation at the 8 th day of pregnancy in the left and right uterus of the experimental group was(1.402±0.517),and(6.420±1.075),respectively.The range of the embryos implanted number in the left uterus of the control group was(6.500±1.090).The range of the right uterine embryo implantation number in the control group was(6.200±0.919).The result revealed that the left embryo implantation number of the experimental group was sig
