摘  要:目的探讨新生猪缺氧缺血性(HI)脑损伤后脑内髓鞘碱性蛋白(MBP)、髓鞘少突胶质细胞糖蛋白(MOG)、髓鞘相关糖蛋白(MAG)在脑白质中表达的变化趋势。方法将28头3~5日龄新生猪随机为对照组(n=4)和HI模型组(n=24)。根据HI后的存活时间,将HI模型组分为6个亚组(n=4),分别为0~2 h、2~6 h、6~12 h、12~24 h、24~48 h、48~72 h亚组。采用免疫组化染色比较各组MBP、MOG、MAG含量的变化。结果HI后MBP的表达整体呈先降低后升高的趋势,6~12 h达到最低值,而后升高,24~48 h后再次降低;与对照组比较,HI模型组0~2 h、2~6 h、6~12 h、12~24 h、24~48 h、48~72 h亚组MBP的表达水平均降低,差异均有统计学意义(P<0.05)。HI后MOG的表达整体呈先升高再降低再升高的趋势,6~12 h达到最高值,而后降低,24~48 h后再次升高;与HI模型组6~12 h亚组相比,HI模型组0~2 h、2~6 h、12~24 h、24~48 h、48~72 h亚组及对照组MOG的表达水平均降低,差异均有统计学意义(P<0.05)。HI后MAG的表达整体呈先降低后升高再降低的趋势,12~24 h达到最低值,而后升高,24~48 h后再次降低;与对照组比较,HI模型组0~2 h、2~6 h、6~12 h、12~24 h、24~48 h、48~72 h亚组MAG的表达水平均降低,差异均有统计学意义(P<0.05)。结论HI后MBP、MOG、MAG的表达变化提示HI脑损伤可引起髓鞘和少突胶质细胞损伤,少突胶质细胞损伤后会激活脑内自发性髓鞘再生机制。Objective To investigate the expression of myelin basic protein(MBP),myelin oligodendrocyte glycoprotein(MOG),and myelin-associated glycoprotein(MAG)in white matter after hypoxic-ischemic(HI)brain injury.Methods A total of 28 neonatal piglets aged 3-5 days were randomly divided into control(n=4)and HI model groups(n=24).The HI model group was further divided into six subgroups based on survival time after HI(0-2,2-6,6-12,12-24,24-48,and 48-72 h).Immunohistochemical staining was performed to detect the contents of MBP,MOG,and MAG in each group.Results The MBP content decreased at first and then increased,reaching the lowest point at 6-12 h,then increased,and decreased again after 24-48 h.Compared with the control group,the MBP content decreased significantly in the 0-2,2-6,6-12,12-24,24-48,and 48-72 h subgroups(P<0.05).The MOG content increased at first,peaking at 6-12 h,following by decrease and subsequent increase after 24-48 h.Compared with the 6-12 h subgroup,the MOG content in the control group and 0-2,2-6,6-12,12-24,24-48,and 48-72 h subgroups significantly increased(P<0.05).The MAG content decreased,following by increase and subsequent decrease after 24-48 h.Compared with the control group,the MAG content significantly decreased in the 0-2,2-6,6-12,12-24,24-48,and 48-72 h subgroups(P<0.05).Conclusion The changes in the expression of MBP,MOG,and MAG after HI suggest that HI brain injury can damage the myelin sheath and oligodendrocytes,and oligodendrocyte injury can activate the mechanism underlying spontaneous myelin regeneration.
