摘　　要：目的探讨长链非编码RNA(lncRNA)MCF2L-AS1在胃癌中的表达情况以及MCF2L-AS1对胃癌增殖侵袭迁移的影响。方法生物信息学分析MCF2L-AS1在胃癌组织中的表达情况;实时PCR检测MCF2L-AS1在胃癌细胞中的表达情况。siRNA转染敲低胃癌细胞中的MCF2L-AS1,实时PCR检测敲低的结果。采用CCK-8试剂检测转染前后胃癌细胞的增殖情况。Transwell实验检测胃癌细胞转染前后的侵袭和迁移能力。富集分析推测MCF2L-AS1可能参与的生物学过程。结果相对于非癌胃细胞和癌旁非癌胃组织,MCF2L-AS1在胃癌细胞和组织中过表达(P<0.05)。MCF2L-AS1敲低后,胃癌细胞的增殖能力减弱,侵袭和迁移能力受到抑制(P<0.05)。MCF2L-AS1可能与细胞周期等生物学功能密切相关。结论lncRNA MCF2L-AS1在胃癌细胞与组织中高表达,并促进胃癌细胞的增殖、侵袭和迁移。Objective To explore the expression of long noncoding RNA(lncRNA)MCF2L-AS1 in gastric cancer and the influence of MCF2L-AS1 on the proliferation,invasion,and migration of gastric cancer.Methods Bioinformatics analysis was performed to assess the expression of MCF2L-AS1 in gastric cancer tissues.The expression of MCF2L-AS1 in gastric cancer cells was detected by RT-PCR.MCF2L-AS1 in gastric cancer cells was knocked down by siRNA transfection and verified by RT-PCR.The CCK-8 reagent and assay were used to detect the proliferation of gastric cancer cells before and after transfection.Transwell analysis detect the invasion and migration of gastric cancer cells before and after transfection.The biological processes that MCF2L-AS1 may participate in were analyzed by enrichment.Results Compared with normal gastric mucosal cells and noncancerous gastric tissues adjacent to cancer,lncRNA MCF2L-AS1 was significantly overexpressed in gastric cancer cells and tissues(P<0.05).After knockdown of MCF2L-AS1,the proliferation of gastric cancer cells was impeded,and the invasion and migration were inhibited(P<0.05).MCF2L-AS1 may be closely related to cell cycle and other biological functions.Conclusion lncRNA MCF2L-AS1 is highly expressed in gastric cancer cells and tissues,and promotes the proliferation,invasion,and migration of gastric cancer cells.
